ObjectiveIn this study,we observed the effects of acupuncture on GnRH and its receptor signal pathway AC/PKA molecules in hypothalamic of tamoxifen-induced hot flashes rat,in order to explore the central mechanism of acupuncture in improving the hot flashes induced by tamoxifen,an endocrine drug after breast cancer surgery.Method42 clean SD female rats were selected and 6 rats with basal body temperature greater than 38 degrees were excluded.The remaining 36 rats were randomly divided into control group?12 rats?and model group?24 rats?.The hot flash model was established by intragastric administration of tamoxifen.After successful modeling,24rats were randomly divided into model group?12 rats?and acupuncture group?12 rats?.The acupuncture group selected"Sanyinjiao","Guanyuan"and"Tai Chong"to carry out the intervention of the manipulation method.Among them,"Sanyinjiao"and"Tai Chong"are altenated in left and right feet.The needles were used to even reinforcing-reducing method.The needle was rotated once every 10 minutes for 1 minute each time.The acupuncture intervention 30 min,once every other day,a total of 10 treatments,lasting 20 days.The temperature of the tail was continuously monitored by a thermocouple thermometer.The serum estradiol?E2?,gonadotropin-releasing hormone?GnRH?and its receptor?GnRHR?were measured by Enzyme Linked Immunosorbent Assay?ELISA?.The expression of adenylate cyclase?AC?and protein kinase A?PKA?in rat hypothalamus was detected by real-time fluorescence quantitative?RT-qPCR?.Results1?The effect of acupuncture on the tail temperature of tamoxifen-induced hot flushes model rats?1?After 28 days of tamoxifen administration,the temperature of the model tail was significantly higher than that of the control group,and the difference was statistically significant?P<0.05?,indicating successful modeling.?2?After 20 days of acupuncture treatment,the tail temperature of the acupuncture group was significantly lower than that of the model group,and the difference was statistically significant?P<0.05?.2?Effect of acupuncture on serum E2 content in hot flushes model ratsCompared with the control group,the serum E2 content in the model group was decreased,while the serum E2 content in the acupuncture group was higher than that in the model group,and approached the control group,but there was no statistical difference between the groups?P>0.05?.3?Regulation of acupuncture on the hypothalamic GnRH and its receptor signaling pathway AC/PKA molecules in tamoxifen-induced hot flushes model rats?1?GnRH concentration in hypothalamus:The concentration of GnRH in the model group was higher than that in the control group?P<0.05?,and the concentration of GnRH in the acupuncture group was significantly lower than that in the model group?P<0.05?.?2?GnRHR concentration in hypothalamus:Compared with the control group,the concentration of GnRHR in the hypothalamus increased,but there was no significant difference?P>0.05?.The concentration of GnRHR in the acupuncture group was significantly lower than that in the model group?P<0.05?.?3?AC gene expression in the hypothalamus:The AC gene expression model group was higher than the control group and the acupuncture group was lower than the model group and the control group,but there was no statistical difference between the groups?P>0.05?.?4?PKA gene expression in hypothalamus:The expression of PKA gene in the model group was higher than that in the control group and the acupuncture group.The expression of PKA gene in the acupuncture group was close to the control group,but there was no statistical difference between the groups?P>0.05?.Conclusion1?Acupuncture can effectively improve the hot flash caused by tamoxifen,and can increase the estrogen content of hot flash rats in a certain range.2?Acupuncture may improve the expression of GnRH and its receptors in the hypothalamus,affecting the expression of signaling pathway-related molecules,thereby improving the hot flash caused by tamoxifen,an endocrine drug after breast cancer surgery.
