Objective:1.Study the effect of exercise on the number of black stool cases in functional constipation model mice within 6 hours,and to clarify the improvement effect of exercise on functional constipation;2.Further study the effect of exercise on the content of serotonin and the abundance of Clostridium in colon,and explore the possible mechanism of exercise in improving functional constipation.Methods:40 SPF Kunming mice of about 8 weeks old were selected,weighing(20±2)g,and all of them were males,provided by the Animal Experimental Center of Shanxi Medical University.Before the experiment was started,all mice needed to be fed adaptively for one week.The mice were divided into four groups,as follows: blank group(A),model group(B),positive group(C)and experimental group(D).From 1st to 14 th day: group A were given normal saline to fill the stomach once a day;group B,group C and group D were given compound diphenoxylate suspension to fill the stomach once a day.From15 th to 38 th day: group A and group B were fed normally without intervention;group C were given clostridium butyricum powder(Baole'an)suspension to fill the stomach once a day;group D kept self-running cage exercising regularly for 2 hours every day.The39 th to 41 st day: after corresponding intervention measures for 30 minutes,each group of mice were given ink to fill the stomach,and recorded the number of black stools of each mouse within 6 hours,once a day,three days in total,took the average of three sets of data.The 42 nd day: after corresponding intervention measures for 30 minutes,allmice were broken neck death,which were used to test the content of5-hydroxytryptamine in colon of mice by ELISA kit construction,and which were used to test the abundance of Clostridium in colon of mice by quantitative real-time PCR.Further analysed the correlation among the abundance of Clostridium,the content of5-hydroxytryptamine and the number of black stools in 6 hours,and explored the possible mechanism of it.Results:1.On the 39 th to 41 st day of the experiment,compared with group A,the number of black stools in 6 hours of group B decreased significantly(P < 0.05);compared with group B,the number of black stools in 6 hours of group C and group D increased significantly(P < 0.05).2.On the 42 nd day of the experiment,compared with group A,the content of5-hydroxytryptamine in colon of group B decreased significantly(P < 0.05);compared with group B,the content of 5-hydroxytryptamine in colon of group C and group D increased significantly(P < 0.05).3.Compared with group A,the abundance of Clostridium in colon of group B decreased significantly(P < 0.05);compared with group B,the abundance of Clostridium in colon of group C and group D increased significantly(P < 0.05).4.The number of black stools in 6 hours was negatively correlated with the abundance of Clostridium in colon and the content of 5-hydroxytryptamine in colon.5.The abundance of Clostridium in colon was positively correlated with the content of 5-hydroxytryptamine in colon.Conclusion:1.Exercise can increase the number of black stools in 6 hours in FC model mice,and improve FC;2.The possible mechanism of improving FC by exercise is to increase the 5-HTcontent of colon by increasing the abundance of Clostridium.
