As the most authoritative diagnostic method in the field of medical diagnosis,pathological examination is regarded as the "golden standard" for malignant tumor identification,and also is the main basis for determining the clinical treatment plan.Paraffin pathological section is the main carrier of diseased tissue and is pathological diagnosis.An indispensable part of it.However,the sample processing flow of paraffin pathological sections is long,and the pretreatment steps(dewaxing,hydration,etc.)are extremely complicated and cumbersome,and have a significant influence on the post-processing results.At present,most hospitals and research institutions in China still use manual methods for pre-processing steps,which have poor stability,low reliability,and potential safety risks.Therefore,the realization of the automation of the pretreatment experimental process has become a pivtol issue to be solved in pathological examination.In this thesis,two sets of fully automatic embedded pathological section pretreatment devices based on large flux or microfluidic chips are designed for different application scenarios.The specific contents include:(1)This thesis systematically introduces the background of pathological detection and microfluidic chip,and studies the implementation methods and problems of current pathological section pretreatment experimental operations at domestic and foreign,the thesis also analyzes the development trend of related automation equipment.The research basis and content of this paper are put forward.(2)Aiming at the application requirements of detection institutions dealing with numerous samples(such as large hospitals or testing centers),a fully automatic large-flux pathological section pretreatment device was designed.The device is composed of a liquid path module,a system control module,a reagent control module,a reagent detecting module and a temperature control module.Among them,the liquid path module adopts a new fixed and closed reaction chamber structure,which avoids the leakage of reagents and pollutes the experimental environment;the reagent detection module uses the U-type photoelectric sensor to check the status of various liquid pipelines,this closed-loop control of the reagent state greatly improves the stability of the experiment.(3)Aiming at the application requirements of detection institutions dealing with a small quantity of samples(such as community hospitals or related scientific research institutions),a pathological section pretreatment device based on microfluidic chip was designed.The pathological section pretreatment chip,the core component of the device,was designed and prepared,and the mechanical structure,hardware circuit and software design and implementation of the peripheral control device were completed.The temperature control module of the device adopts a position control strategy with heating time as the control variable,and the heating rate is improved while ensuring that the temperature control accuracy meets the experimental requirements.The adoption of the microfluidic chip greatly improves the utilization rate of expensive reagents and significantly reduces the hardware volume.Finally,the thesis tested two devices by experiment.The results show that the device is stable in operation,simple and convenient to operate,high in automation and reliability,greatly improving the quality and efficiency of the pathological section pretreatment experiment;the device also greatly saves the use of expensive reagents and significantly reduces the production cost.All experimental indicators have reached the expected design goals.
