Eugenics have great practical significance for reducing the probability of congenital malformation newborns,the level of reproductive health,and even improving the overall health level of population.At present,the main clinical diagnosing markers for clinical testing of eugenics are called TORCH,which are made up of five pathogens:toxopasma(TOX),rubella virus(RUB),cytomegalovirus(CMV),and herpessimplxvirus(HSV,which are divided into HSV-I and HSV-II)..Among the existing TORCH detection methods,the detection instrument can only complete the detection of one biomarker in one detection process,which results in a long total detection time of the five indicators,tedious and repetitive detection process,low detection efficiency,high detection cost,and large volume of detection reagent consumption.In order to achieve the multiple diagnosis of five indicators and multiple immune detection at one time.Based on the microfluidic technology,this paper studies a TORCH detection method and device based on the microfluidic chip.With the help of microchannels,microreactors and other functional units integrated in the microfluidic chip,an integrated and automated detection system is built,which significantly simplified the detection process,overcomes manual operations,and improves detection efficiency.In particular,through the automatic sample processing function of microfluidic chip,five multiple immune detection functions of TORCH can be realized,which is useful for various applications It is of great significance to detect eugenics in the field,even point of care testing(POCT).This paper studies the five detection methods of TORCH,designs the matching microfluidic detection chip and integrated detection device.First,this paper studies the microfluidic chip based on the microsphere immunosensor:In order to achieve the TORCH joint inspection,five microsphere immunosensors coated with different antigens are integrated into the microfluidic chip.Thus,five microsphere immunosensor can capture the biomarkers of the sample to be tested simultaneously and achieve multiple immune detection.The reaction reagent is stored in the reagent bottle,and during the reaction,it is gradually released to the detection chip quantitatively.The microfluidic chip realizes various fluid control on the chip,including sample/reagent injection,mixing,waste liquid evacuation and residual waste liquid removal,which are realized by the cooperation between the reagent receiving unit,the microsphere fixing unit,the reaction channel,the waste liquid chamber,and the chip structure.At the same time,this paper studies the supporting testing instruments of the microfluidic chip.Its main functional modules includes:reagent release module,fluid drive module,heating module and image detection module,etc.At the same time,combined with hardware and software design,and even image processing algorithm design,the overall function of the testing instrument is realized.This paper also studies the various influencing factors of the TORCH joint inspection microfluidic device,depending on this basis,the comprehensive performance of the system was further optimized.Through the cooperation of microfluidic chip and supporting detection instruments,five multiple immune tests of TORCH can be completed in 55 minutes.The test results show that its comprehensive performance has basically met the requirements of clinical detection,which lays a solid foundation for the realization of efficient TORCH five multiple immunoassay in the field rapid detection environment.
