Exosomes,which are lipid membrane-enclosed nanovesicles?30-150nm in diameter?,contain a plethora of proteomic and genetic information.Specifically,tumor-derived exosomes have been recognized as ideal biomarkers for cancer diagnosis.However,effective methods for isolating as well as detecting exosomes are still challenging.Herein,we have developed an immunoaffinity-based microfluidic device HBEXO-Chip which can directly separate exosomes from plasma samples.This separation technology enables capturing the exosomes through tumor specific surface markers,such as the pancreatic cancer target Glypican-1?GPC1?employed in this study,thus preventing the interference of other exosomes.Our study demonstrated that pancreatic cancer can be distinguished from liver cancer and healthy control group by the device.There were 81 exosomal miRNAs which were differentially expressed in pancreatic cancer patients as compared to the healthy control group.Moreover,three exosomal miRNAs,which might possess diagnostic value for pancreatic cancer,were found through bioinformatics analysis.Altogether,the microfluidic device HBEXO-Chip could be beneficial to find out exosomes-based biomarkers for differential diagnosis of pancreatic cancers and contributes significantly to the basic research of pancreatic cancer based miRNAs.
