Purpose:The expression of growth differentiation factor-15(GDF-15)in breast cancer and adjacent tissues was examined,and the relationship between GDF-15 and clinicopatho logical features and prognosis of breast cancer was explored..Methods:75 cases of primary invasive breast cancer in the Second Affiliated Hospital of Soochow University were selected between January 2009 to June 2014.30 cases of adjacent paracancerous tissues were selected as controls.Immunohistochemical staining was used to detect the expression of GDF-15 in breast cancer and adjacent tissues.The expression of GDF-15 was analyzed in the tissues,and all follow-up data were statistically analyzed to investigate the correlation between GDF-15 expression and clinicopathologieal features of breast cancer,and to study the effect of differences in expression levels on the outcome of breast cancer patients.Results:The positive expression rate of GDF-15 in breast cancer tissues was significantly higher than that in adjacent tissues,and the positive rates were 52%and 6.67%,respectively,and the difference was statistically significant(P<0.01).The positive rate of GDF-15 expression was significantly different in different axillary lymph node status,histological grade and pathological stage(P<0.05).The more the axillary lymph node metastasis,the higher the histological grade and pathological stage,the higher the positive rate.The positive rate of GDF-15 expression was not related to patient age,menstrual status,tumor diameter and molecular typing.Survival analysis showed that the overall survival rate of GDF-15 positive group was 71.79%,the negative group was 91.67%,the difference was statistically significant(P<0.05);the GDF-15 positive group had a disease-free survival rate of 58.97%,and the negative group was 88.89%,the difference was statistically significant(P<0.01).Multivariate analysis suggested that GDF-15 is a prognostic indicator for breast cancer patients,Patients with positive GDF-15 expression had shorter OS than negative patients,HR=1.97,95%Cl:1.01?3.82,P<0.05;patients with positive GDF-15 expression had shorter DFS compared with negative patients,HR=2.08,95%Cl:1.19 to 3.64,P=0.01.Conclusions:The expression of GDF-15 was significantly increased in breast cancer tissues,and the expression level of GDF-15 was significantly correlated with histological grade,Pathological stage and axillary lymph node metastasis.This study demonstrates that GDF-15 plays an important role in the development of breast eancer.The role is a prognostic indicator for breast cancer survival.Detection of its expression in breast cancer tissues has important reference value for predicting the progression and prognosis of breast cancer.
