Objective: Quality control of Hawthorn medicinal materials with different germplasms and harvesting dates was studied.A multi-component and multi-disciplinary quality control system of Hawthorn pharmacodynamics was established to improve the quality evaluation methods of Hawthorn medicinal materials.Methods: To study the powder microscopic characteristics of Hawthorn in different germplasms and different harvesting periods;to establish a new method for identification of Hawthorn by TLC;to determine the contents of total acids,flavonoids and total sugars(polysaccharides,soluble sugars)in Hawthorn by acid-base titration and colorimetry;to determine the representative chemical constituents of organic acids and flavonoids in Hawthorn in different germplasms and harvesting periods by HPLC.Content determination and fingerprint study.Result: 1.There are obvious differences in appearance,flavor and breakage rate of hawthorn fruit among different germplasms and harvesting periods: Big five-edged Hawthorn has big head,five-edged protuberance and sweet red seed odor;Big Venus Hawthorn peel gradually changes from green to red,single fruit weight from 4g to 12 g,single fruit weight from 8g to 14 g,breakage rate from 6.17% to 43.01% in different harvesting periods.2.The microscopic characteristics of hawthorn powder in different germplasms and harvesting stages are different: the pigments in sweet red seed parenchyma cells are bright red,the big pentagonal starch granules are obvious,the threaded vessels are more,the wild hawthorn stone cells have two corresponding sections,and the cell walls and cell cavities of big Venus and big asbestos ball Hawthorn are different in different harvesting stages,which can be used as the qualitative and identification basis of hawthorn.3.The contents of total acid,total flavonoids,total sugar(soluble sugar,polysaccharide)of Hawthorn in different germplasms and harvesting stages were significantly different.The content of total sugar in Sweet Red Hawthorn was higher than that in Dajinxing,which could be used as the qualitative and identification basis ofHawthorn.4.A new TLC method for identifying flavonoids such as rutin,Hyperoside and isoquercetin in hawthorn fruits was supplemented,which realized the comprehensiveness and accuracy of identifying different hawthorn germplasms,and could be used as qualitative basis for hawthorn harvest time.5.The content of organic acids and flavonoids in Hawthorn was determined by HPLC and the fingerprints were studied.It was found that there were differences in the content of chemical components in different hawthorn germplasms,and the transformation degree was small when the harvest time was the same month.Among them,the catechins of Big Venus,Small Venus and Big Five Ridges were higher,while wild hawthorn and hawthorn rutin were lower.The pharmacodynamic components of Hawthorn were evaluated comprehensively and concretely.Conclusion: Hawthorn medicinal materials of different germplasms and different harvesting periods were systematically studied at different levels and in all directions.A multi-disciplinary identification and content determination system for the main components of hawthorn was established.The evaluation method of quality standard of Hawthorn medicinal materials was improved,which provided scientific data support for the development and utilization of Hawthorn germplasm resources.
