Rhubarb is a genuine medicinal material of Gansu Province, and its chemical compositions include anthraquinones, stilbene glycosides, chromones, naphthol glycosides and tannins. Rheum tanguticum was included in Chinese pharmacopoeia(2010 edition). In order to guarantee the resources advantages and accelerate the cultivation-bases construction of Rheum tanguticum in Gansu Province, GAP cultivation-bases of Rheum tanguticum were built in Tibetan Autonomous Prefecture of Ganan. In this paper, the following 3 contents were systematically researched:A Determining the optimal harvest time of Rheum tanguticumFive anthraquinones compositions in Rheum tanguticum were determined by HPLC, and the samples were from Zhagulu Town, Tibetan Autonomous Prefecture of Ganan and harvested in different growth periods from three to four years. The sum content of five anthraquinones compositions in Rheum tanguticum harvested in September and October of the fourth year were 4.30% and 5.13%, and were higher than that harvested in other months.So the month of October of the fourth year could be determined as the optimal harvest time of Rheum tanguticum.B Research on the quality standards of Rheum tanguticumA enterprise quality standard of GAP-cultivation in Tibetan Autonomous Prefecture of Ganan was built based on related contents in Chinese pharmacopoeia(2010 edition). The temporary standard of Rheum tanguticum included the flowing contents: moisture content ≤12.0%, total ashes ≤ 10.0%, acid-insoluble ashes ≤ 0.8%, Cu ≤ 20.0mg/kg, Cr ≤ 0.3mg/kg, Pb≤ 5.0mg/kg, Cd ≤ 0.3 mg/kg, water extract > 25%, ethanolic extract > 15%, and content determination > 1.5%.The study of HPLC fingerprint at the wave of 270 nm of 10 batches of Rheum tanguticum showed that the similarity were between 0.710 ~ 0.911, and 51 peaks in the fingerprint were confirmed while 10 effective components(sennoide A, sennoide B,chrysophanol-8-O-β-D-glycosidase, emodin-8-O-β-D-glycosidase, physcion-8-O-β-D-glycosidase, aloe-emodin, rhein, emodin, chrysophanol, and physcion) were defined by reference substances. The results of PCA(principal component analysis) and HCA(cluster analysis)showed that the 10 samples were classified into 2 groups, the samples from Tibetan Autonomous Prefecture of Gannan and Qinghai Province were classified into one group, and from Sichuan Province were another. The results of PCA showed that emodin, chrysophanol,and physcion were more important components in Rheum tanguticum.C Research on pharmacodynamics of Rheum tanguticumThe research on purgative effect of Rheum tanguticum showed: Compared with the model group, the difference of the first time of passing black stool and its frequency within 5h after administration of three kinds of Rhubarb showed statistically significant(P <0.05).Rheum tanguticum from Gannan group and Xining group on enhancing the wet weight of intestinal, water content of intestinal and intestinum crassum was stronger than Lixian group(P <0.01). Purgative effect of Rheum tanguticum from Gannan group showed no significant difference with that from Xining group, but better than Lixian group.The research on anti-inflammatory effect of Rheum tanguticum showed: Compared with the normal control group, the mice ear swelling and rat paw swelling after administration of three kinds of Rhubarb showed statistically significant(P <0.05). Compared with the model group, the content of total proteinin exudate of mice balloon synovitis and LTB4 in three kinds of Rhubarb group showed statistically significant(P <0.01). Anti-inflammatory effect of Rheum tanguticum from Gannan group showed no significant difference with that from Xining group, but better than Lixian group.The research on Lowering blood pressure, improving blood rheology and antioxidant effect of Rheum tanguticum showed: Compared with the normal control group, the content of TC, TG and LDL-C increased while the content of HDL-C and MDA decreased(P<0.05).Compared with the model group, the blood viscosity of hyperlipidemia rats generally decreased(P <0.05).The effect of Lowering blood pressure, improving blood rheology and antioxidant of Rheum tanguticum from Gannan group showed no significant difference with that from Xining group.
