Objectives: The relationship between the expression of miR-16 in hela cells and the sensitivity of cisplatin was discussed.Methods : Lipofectamine2000 mediated miR-16 mature analogue(mimics)and negative control fragments(NC)were transfected with cervical hela cells.Real-time fluorescence quantitative PCR was used to detect miR-16 expression in each cell.The expression of Bcl-2 and Bax?caspases-3 protein in cells is detected by protein imprinting.The survival rate of cisplatin cells is detected by tetramethylazolazole blue(MTT)colorimetric method.Result:1.MiR-16 transfection is highly expressed in Hela cells2.MTT results shows that miR-16 overexpression significantly increased the sensitivity of hela cells to cisplatin.3.Western blot results indicate that,compared with the control group,Hela cells of miR-16 in overexpression group,the expression level of Bcl-2 was elevated,and the expression level of Bax and caspase-3 decreased.Conclusion:MiR-16 can enhance of cervical cancer hela cisplatin sensitivity,miR-16 may be targeted by cervical cancer hela Bcl-2 protein expression,raised Bax and caspase-3 protein expression,and provide new targets for the treatment of cervical cancer.
