Apoptosis And Expression Of Caspase-3 In Cochlea Hair Cell After Cisplatin Toxicity

ObjectiveTo investigate the character of damaged cochlea hair cells caused by cisplatin, and to identity whether apoptosis and Caspase-3 occur in the damaged cells.MethodHealthy guinea pigs were divided into experimental group and control group randomly with 10 of each, animals in experimental and control group received systemic injection of cisplatin (2mg·kg-1·d-1) and saline with the same dose respectively for 6 consequent days. DPOAE was applied to each guinea pigs before and after 6 days that cisplatin was injected. All the animals were sacrificed on the 7th day, the cochlea of each was observed by frozen dissection. The apoptosis of hair cells were detected by TUNEL method and Caspase-3 were detected by immunochemistry.
