Objective:To study Cisplatin combined with Tiji’ao in vitro anti-tumor effect on human gastric cancer MGC-803cell line, and its mechanism exploration preliminary to discuss that Cisplatin combined with Tiji’ao used clinically to provide the experimental basis and theoretical basis.Method:First using MTT into preliminary test, the detection of Cisplatin combined with Tiji’ao in vitro human gastric cancer MGC-803cell line anti-tumor effect; then morphology was observed the main method inverted microscope, and smear of cell HE staining and transmission electron microscopy observation of gastric cancer MGC-803; cycle distribution and apoptosis rate of detection method for the combination of two drugs using flow cytometry The role of the results of the MGC-803gastric cancer; mechanism for the use of immunohistochemical staining, detection of Caspase-3, caspase-8and Caspase-9three proteins’ expression,depth study Cisplatin combined with Tiji’ao gastric cancer MGC-803cell lines induced apoptosis.Results:(1) tetrazolium salt (MTT) assay results showed that:Cisplatin combined with Tiji’ao has anti-value-added effects on gastric cancer MGC-803cell lines.the inhibition rate and the dose was positively correlated with increasing drug concentrations inhibit the rate was also an upward trend (P<0.01);(2) HE staining results showed that:Cisplatin combined with Tiji’ao a final concentration of20μg/ml role in gastric cancer MGC-803cells apoptosis, after that morphological changes,showing that part of the cell formation of apoptotic bodies. Cells visible nuclear condensation, nuclear fragmentation and karyolysis and apoptosis performance;(3) inverted microscope results showed that:Cisplatin combined with Tiji’ao20μg/ml role of cells apoptotic,after that morphological changes:cells lose the original polygon structure into a round apoptotic cells; even some cells completely broken, the dead cells, was in the microscopic debris performance;(4) TEM observation showed that:Cisplatin combined with Tiji’ao20μg/ml role in gastric cancer MGC-803cells, the nuclear chromatin gathered into a massive structure, visible chromatin part of the fracture performance, can be the formation of apoptotic bodies;(5) Flow cytometry results further show that:Cisplatin combined with Tiji’ao can be induced gastric cancer MGC-803, apoptosis, cell cycle protein test results showed GO/G1of the ratio was significantly increased (P<0.01), the decline in the ratio of the S and G2/M phase, indicating that cell cycle arrest in GO/G1period;(6) immunocytochemistry mechanism show that:cisplatin combined with Tiji’ao and the control group (PBS), compared to caspase-3, caspase-8and Caspase-9expression showed an increasing trend, the results are obvious(P<0.01).Conclusion:1, Cisplatin combined with Tiji’ao gastric cancer MGC-803cell lines with anti-proliferative effect.2, due to the emergence of a large number of apoptotic bodies or apoptotic cells, can prove that Cisplatin combined with Tiji’ao has anti-proliferative effect on gastric cancer MGC-803, when it in20μg/ml mostly leads to apoptosis.3,Cisplatin combined with Tiji’ao has effects on gastric cancer MGC-803cell lines, inhibition of proliferation and induction of apoptosis, the mechanism based on the experimental proof of the main could be suppressed by the application of Cisplatin and Tiji’ao in the cells in GO/G1phase and can organize this on the cells cycle; these two drugs in combination can enhance the caspase-8, spase-9and Caspase-3protein expression, may initiate apoptosis, and the implementation of the death receptor pathway.
