Objective:Purpose one:To study the prescription regularity by Data mining on the application of herbs in the prescriptions of Chinese herbs for the treatment knee osteoarthritis.Purpose two:To observe the effect of monomer of Chinese traditional herbs which was the most widely-used herbs selected by data mining combined with BMSCs on the repairing cartilage defect.MethodsMethod one:Screening in the prescriptions of Chinese herbs for the treatment knee osteoarthritis in CNKI,Wanfang and VIP database before December 2017.SPSS software and data mining method were used to explore the formulations for the treatment of knee osteoarthritis with high frequency drugs,drug pairs and core combinations.Method two:The best concentration and action time of effective Chinese medicine monomer of the screened Chinese medicine was measured by CCK-8 assay.After the model of articular cartilage defect in the knee was constructed,healthy New Zealand white rabbits were randomly divided into model group,Chinese medicine monomer group,BMSCs group,Chinese medicine monomer+BMSCs group.Samples were taken and histological score of ICRS was obtained after intraarticular injection of BMSCs for 4 weeks.HE staining and toluidine blue staining were used to observe the repair effect.ResultsResult one:140 literatures which meet the inclusion criteria,140 prescriptions and 217 Chinese medicines was included.10 herbs with high requency among them were twotoothed achyranthes root,herba epimedii,angelica sinensis,radix rehmanniae praeparata,radix polygoni alba,eucommia ulmoides,caulis spatholobi,drynaria rhizome,astragalus membranaceus and ligusticum wallichii.Combined with modern basic research,icariin,the effective components of herba epimedii,were screened and applied to intervene BMSCs.We explored the compatible regularity of traditional Chinese medicine in treating knee osteoarthritis through factor analysis of the top 20 Chinese medicines in frequency and concluded 7 pairs of factors,including loranthus parasiticus and radix angelicae pubescentis;cassia twig and radix polygoni alba;fructus psoraleae and caulis spatholobi;astragalus membranaceus and angelica sinensis;atractylodes and poria cocos;radix rehmanniae praeparata and eucommia ulmoides;pawpaw and drynaria rhizome.We summarized the core ombinations of Chinese herbs through association rules analysis,including twotoothed achyranthes root,eucommia ulmoides and radix rehmanniae praeparata;angelica sinensis,eucommia ulmoides and radix polygoni alba;herba epimedii,caulis spatholobi and radix rehmanniae praeparata;twotoothed achyranthes root,eucommia ulmoides,radix polygoni alba and angelica sinensis.Result two CCK-8 assays showed that icariin could enhance the activity of BMSCs,with the best concentration of luM and the best intervention time of 72h.After 4 weeks of modeling,we found that the size of cartilage defect in the group which treated BMSCs with icariin was significantly reduced than others groups,and ICRS histological score was higher than other groups(P<0.05).HE staining and toluidine blue staining suggested that BMSCs combined with icariin showed better repairing effect than BMSCs and the repairing tissue was well-integrated with adjacent tissues.ConelusionConclusion one Through data mining analysis,the commonly used traditional Chinese medicines in treating knee osteoarthritis were twotoothed achyranthes root,herba epimedii and the commonly used Chinese traditional medicine partner loranthus parasiticus and radix angelicae pubescentis.The data mining is helpful to summarize the rule of Chinese medicine prescription and some use for reference for clinical medication.
