Depression is an emotional disorder that seriously affects the normal life of patients.Patients with depression often fall into emotions of grief,inferiority and pessimism.At the same time,the somatization disorder will also generate the physical pain for patients and it also increases a lot of medical burden and economic costs.In addition to creating a double threat to patient's mental and physical,depression also have many complex interaction relationship with other diseases.Therefore,it is of great significance to explore appropriate control and treatment strategies for depression.Jieyu Pill(JYP)is an antidepressant formula based on Traditional Chinese Medicine theory and formed in long-term practice,many experiments and clinical studies have proved that it has good antidepressant effect.Traditional Chinese Medicine has different ideas with biomedicine in treating depression.Through the analysis of the mechanism of effective Traditional Chinese Medicine formulas,it can help to provide new ideas for biomedical understanding and treatment of depression.The current research on JYP is mainly focused on the observation of clinical efficacy,and the combination application with other drugs as well as the comparison of efficacy,but there are very few studies on the pharmacological mechanism.This is because Traditional Chinese Medicine has a variety of components,and it will take a lot of time,manpower and material resources to conduct pharmacological experiments and verify efficacy one by one,and due to the lack of basic data support,it is impossible to start when selecting components and genes for research,and it is impossible to fully understand the pharmacological mechanism of JYP.The development of system science and informatics has made it possible to study under such conditions.It is possible to study the mechanism of Traditional Chinese Medicine on the basis of integrating the existing knowledge of biology,and discover the characteristics of Traditional Chinese Medicine in treating diseases on the basis of mechanism analysis.In summary,this study will explore the appropriate treatment strategies for depression based on the systematic analysis of the pharmacological mechanism of JYP by using informatics methods.In order to systematically analyze the pharmacological mechanism of JYP,study the effects of somatization disorder on the progression of depression,and provide feasible treatment strategies for depression,this paper has conducted a series of studies,including the four aspects:(1)By integrating all the information of the chemical components of JYP,the target protein of chemical components,protein-protein interaction data,depression-related genes,144 types of cells,genes expressed in cells,ligands and receptors on cells,ligand-receptor pairs.In this paper,Neo4j is used to construct a graph database for the analysis of the pharmacological mechanism of JYP,and as a query platform and research platform for background data and related information in this study.(2)Setting up rules for data screening and using the entity grammar system to construct the mechanism network of JYP,this paper will analyze the chemical components that play a key antidepressant role and the key genes related to depression,and explore the signal pathways where the key genes are located as well as its relationship with the development of depression.(3)Using cell communication data,the key chemical components and key genes to analyze the transfer of antidepressant activity of JYP in cells and the function it main impacted,this paper from a more macro level analyzes JYP how to exert antidepression effect by comprehensive regulation of body function.(4)The influence of genes related to insomnia and climacteric syndrome on the occurrence and development of depression was analyzed,and the mechanism of JYP treatment of these two somatization symptoms was analyzed using the entity grammar system.The construction of graph database offers support for systematically analyzing the pharmacological mechanism of JYP.The mechanism network intuitively shows the relationship between the chemical components of JYP and disease-related genes.In the analysis of key nodes,15 key chemical components that play a role and 20 key depression-related genes are obtained,which provides basic data such as lead compounds and intervenable target genes for the depression treatment.The analysis of the signal pathways where the key genes are located indicates that JYP play a comprehensive regulatory role by acting on multiple biological processes related to depression.The study of antidepressant activity in cell-to-cell transmission displays JYP mainly act on immune cells and nerve cells,and possess a certain regulatory effect on the body's digestive function,vascular function and hematopoietic function,which represents that interfering with immune cells and improving the body's immunity is a new thinking direction for the treatment of depression.The research results on the relationship between somatization symptoms and depression show that the somatization disorder affect the procession of depression,which indicates that intervention of somatization disorder is also a key issue to be considered in the study of depression treatment strategy.This study provides key chemical components that can be further developed,the interferable target genes,cell types,and some somatic symptoms for the treatment of depression.In addition,this study systematically analyzes the pharmacological mechanism of JYP and integrates it into the Neo4j graph database,providing a systematically and efficiently analysis method and platform.This study expands the clinical application of JYP and provides basic data for the further development of it.Depression is a complex disease,the biomedical interventions and treatments that focus on the nervous system have obvious shortcomings in clinical efficacy,so how to use drugs to treat depression requires in-depth discussion.This study puts forward different ideas for the treatment of depression based on the systematic analysis of the mechanism of clinically effective prescription JYP.It mainly includes the following two aspects:(1)Depression is a disease in which the pathological state of the mental system and the somatic symptoms are mutually causal and mutually promoting,therefore,the treatment strategy should consider the simultaneous treatment of two aspects in order to block the continuous development of depression;(2)Depression is a systemic disease that involves many different cell types and cell functions.The treatment strategy should be designed on the basis of a systematic analysis of the relationship between related cells.Through the study of the mechanism of JYP in treating depression,this study also explored a feasible technical approach for biomedicine to learn from the principles of Traditional Chinese Medicine.
