Objective:To observe the effect of warming yang and supplementing kidney moxibustion therapy in improving VD rats cognitive function.To observe its effection on NF-kB signal pathway and analysis its possible mechanismMethod:1.SD rats were randomly divided into sham operation group,model group,moxibustion group and western medicaton group,with 10 rats in each group2.To verify the effect of warming yang and supplementing kidney moxibustion therapy in improving cognitive function compaired with western medication group,SD rats were established into VD models by appiying the ischemia reperfusion method in bilateral commom carotid arteries3.Observe cell ultrastructure by electron microscope;western blot analysis were performed to detected the expression level of hippocampal IL-1??TNF-??p-I?B?NF-Kbp65?NF-kBp504.RT-qPCR were performed to detected the expression level of IL-1??TNF-??NF-Kbp65?NF-kBp50 mRNA in each group,and reveal its mechanism on restrain inflammatory reactionResults:1.Following modeling,the average escape latency of Morris maze water swimming tests was significantly increased compared with the sham group(P<0.05).After treatment,the moxibustion group did not show significant increase compared with the model group2.After treatment,the increased TNF-??p-I?B?NF-?Bp65 protein3.After treatment,IL-1??TNF-??NF-?Bp65 mRNA were reversed in both moxibustion and medication groups compared with the modle group(P<0.05)4.The model group show a loosely distributed and pyknotic nuclei in hematoxylin and eosin stain,an increasing number of damaged neurons and vague structure and a deeply eosinophilic cytoplasm,while the moxibustion and medication group show a relatively less eosinophils infiltration and damaged neuronsConclusion:Moxibustion is effective in treating VD,which may be association with its functions in down-regulating the levels of IL-1??TNF-? and NF-?B related gene,supressing NF-?B Signaling Pathway and inflammatory cascade. |
