Objective:To observe the effect of huayu tongluo moxibustion on neurovascular units in brain of VD model rats and to explore the mechanism of huayu tongluo moxibustion on vascular dementia.Method:The sham operation group was randomly divided into 15 rats,VD animal models were prepared by modified two-vessel occlusion(2-VO),after successful model identification,the rats were randomly divided into model group,moxibustion group and western medicine group,15 rats in each group.Moxibustion group to remove huayu tongluo moxibustion method for treatment,everyday for each point for 20 min,a course of treatment for a week.The western medicine group was intraperitoneally injected with rat nerve growth factor at a dose of 1.8?g/kg.Both groups were treated 7 times a week,one week as a course of treatment.Morris water maze was used to detect the effect of resolving blood stasis and tongluo moxibustion on VD rats' spatial memory after three courses of treatment.The rats were divided into the sham operation group,the model group,the moxibustion group and the western medicine group by the same molding and grouping method.Immunohistochemical and Western blot method to detect different time points within the VD rats hippocampal the proteins expression of Nestin,DCX,CD34,Collagen IV.Result:1)Morris water maze test the behavioral results of rats after three courses of treatment showed that:Compared with the sham operation group,there was no significant difference inswimming speed among the model group,moxibustion group and western medicine group(P>0.05).The mean value of four days' escape latency in the model group was prolonged,and the difference was statistically significant(P<0.01).The number of rats crossing the platform in the model group decreased(P<0.01).The first crossing time of the model group was significantly prolonged(P<0.01),and the first crossing time of the moxibustion group and the western medicine group was significantly prolonged(P<0.05).Compared with the model group,the average escape latency of moxibustion group and western medicine group was significantly shortened(P<0.01).The number of crossing the platform in moxibustion group and western medicine group increased,with statistical significance(P<0.01?P<0.05).The first crossing time of moxibustion group and western medicine group was shortened(P<0.05).Compared with the western medicine group,there was no significant difference in the average value of the escape latency,the number of crossing the platform,and the time of the first crossing the platform between the moxibustion group and the western medicine group(P>0.05).2)Expression of neurovascular unit related proteins:Immunohistochemistry and Western blot results showed that at the end of a course of treatment,the expressions of Nestin,DCX and CD34 in the moxibustion group and Western medicine group were significantly higher than those in the sham operation group and the model group,and the differences were statistically significant(P<0.01).At the end of the two treatment courses,the expressions of Nestin,CD34 moxibustion group and western medicine group reached the peak.At the end of three courses,the expression of DCX moxibustion group and western medicine group reached the peak.Collagen IV at the end of one period of treatment,reduced compared with control group,model group expression(P<0.01),of which the Wb shows that compared with model group,moxibustion group and western medicine group were significantly increased,the difference was statistically significant(P<0.01).At the end of the three cycles,CollagenIV moxibustion group and western medicine group expression to the peak.The results showed that the huayu tongluo moxibustion could significantly stimulate Nestin,DCX,CD34,the expression of Collagen IV proteins.Conclusion:1)Huayu tongluo moxibustion can improve the spatial memory ability of VD rats.2)Huayu tongluo moxibustion may up-regulate the expression of Nestin and DCX by regulating the relevant proteins of neurovascular units,so as to promote the proliferation,differentiation and migration of neurons.The expression of CD34 was up-regulated to assist angiogenesis.Increase Collagen IV express strong blood brain barrier.Furthermore,it can stabilize the balance of the whole neurovascular unit and improve the VD spatial memory dysfunction.
