Background:Nesfatin-1 was a newly discovered polypeptide with anti-feeding effect.It was derived from nucleobindin-2 and was widely distributed in central and peripheral tissues.The higher expression sites mainly included the feeding regulation center of the hypothalamus,stomach,pancreas,etc.Nesfatin-1 had a variety of important physiological regulatory functions,including regulating food intake,maintaining energy homeostasis,and regulating stress and mood,etc.The effect of age on Nesfatin-1 had not been reported yet.In this study,we demonstrated the expression of Nesfatin-1 in serum ofdifferent age groups and its correlation with other clinical indicators.Objective:To analyze the levels of expression of Nesfatin-1 in serum of different age groups and their correlation with other clinical indicators,and to explore the trend of serum Nesfatin-1 levels with age.Methods:The healthy population who participated in the physical examination at the Physical Examination Center of the Second People's Hospital of Wuhu from January2018 to December 2018 was selected.The general basic data including the age,sex,height and weight was calculated through questionnaire survey,calculating body mass index BMI,BMI=weight/height~2(kg/m~2).The subjects were also tested for fasting blood glucose FPG(hexose kinase method),total cholesterol TC(enzymatic method),triglyceride TG(enzymatic method)and thyrotropin TSH(chemiluminescence method)results.Serum Nesfatin-1 levels of all subjects were measured by enzyme-linked immunosorbent assay(ELISA),and analyzing serum Nesfatin-1 levels in different age groups and their association with age,FPG,TC,TG and TSH.Results:1.The healthy population was divided into three groups according to age:young group,middle-aged group and old group.Serum Nesfatin-1 levels of these three groups were 731.6±82.2pg/ml,769.8±127.8pg/ml,1015.4±87.7pg/ml.The serum Nesfatin-1 level in the elderly group was significantly higher than that in the young group and the middle-aged group(P<0.05),the difference was statistically significant.There was no significant difference between the young group and the middle-aged group(P>0.05).2.Pearson correlation analysis showed that serum Nesfatin-1 levels were positively correlated with age(r=0.614),FPG(r=0.519),TC(r=0.438),and TSH(r=0.499)in healthy people(P<0.05).3.Nesfatin-1 was the dependent variable,and other clinical indicators were independent variables.Multiple linear regression analysis showed that age(?=0.346,P<0.05),FPG(?=0.188,P<0.05),TSH(?=0.187,P<0.05)were the influencing factors of serum Nesfatin-1 levels.4.In healthy people,the serum levels of Nesfatin-1 in male and female of the young,middle-aged,and elderly group were 726.5±159.7pg/ml,755.8±144.1pg/ml;751.7±142.5pg/ml,800.4±104.0pg/ml;987.4±113.5pg/ml,1022.3±131.5pg/ml,there was no significant difference betweenmale and female in the three groups.(P>0.05).Conclusion:Serum Nesfatin-1 levels gradually increase with age in healthy people,and serum Nesfatin-1 levels in the elderly group significantly increase;age,FPG and TSH are the influencing factors of serum Nesfatin-1 levels;There is no gender difference in serum Nesfatin-1 levels in healthy people.
