Study On Quality Characterization Of Single Herbs Of Huangqi Guizhi Wuwu Decoction And Gancao Xiexin Decoction Standard Granules

In order to overcome the shortcomings of traditional Chinese medicine compound decoction,such as inconvenience of decocting and carrying,difficulty in definiting a common criterion of the compound decoction.Chinese medicine standarded granules has been developed.The basically consistent of standarded granules and compound decoction which based on the material,is the key to promote the research project.The quality study of the standarded granules is on the premise that quality study of the decoction referring to the traditional compound decoction.At the early stage,our research group had already accomplished the quality characterization study of the Standard Decoction of Huangqi Guizhi Wuwu Decoction and Gancao Xiexin Decoction.On this basis,we will get a further study on the quality characterization of every single herb that used in each prescription,connect the quality of standarded granules with the quality of the herbs which mentioned in the prescription,and provide scientific data and methods for the quality study of the final drug product accordingly.Huangqi Guizhi Wuwu Decoction is from the Synopsis of Prescriptions of the Golden Chamber,make up of 5 traditional Chinese herbs,astragalus,cassia twig,radix paeoniae alba,ginger and jujube.Gancao Xiexin Decoction,a famous prescription on the Treatise on Febrile Disease,uses 7 kinds of traditional Chinese herbs,licorice root,pinellia,radix scutellariae,rhizoma coptidis,ginseng,dried ginger,jujube.According to the general requirements of the project,our research group have already finished the preparation and quality characterization study of those two compound decoctions at the early stage.The work can be divided into three parts:the determination of the rate of water decoction,the establishment of the fingerprint chromatograms of the chemical composition,the study on the determination of Index Component Content.We have finished the paste rate determination of 7 single herbs,including the whole five herbs in Huangqi Guizhi Wuwu Decoction and the other two in the Gancao Xiexin Decoction,radix scutellariae and rhizoma coptidis.Besides,we have established the HPLC fingerprint chromatograms of the chemical composition of those herbs mentioned above except jujube.Also,we have finished the similarity evaluation of these 6 herbs in 10 different batches which are distinct from different place of origin,and then used HPLC-Q-TOF-MS/MS to receive the qualitative identification of the common chromatographic peaks of 6 herbs' HPLC chemical fingerprint composition.Finally,established the determination methods of the index components of 3 single herbs(radix astragali,radix paeoniae alba and radix scutellariae),determinated the indicator elements transfer rate of those 3 kinds of herbs in 10 different batches.To a certain extent,the study has embodied the potential relevance of a single herb with the reference decoction in the aspect of characteristic component and index component content.This study provides a scientific way to figure out quality characterization and control of every single herb mentioned above,leave useful experience for the quality characterization and control of the reference decoction or granules as well.
