Objective:Explore licorice aconite decoction and huangqi guizhi wuwu tang analgesic effect of cold dampness syndrome pain model mice,based on the nf-kappa B signaling pathway to explore licorice aconite decoction and huangqi guizhi wuwu decoction on haemorrheological nature osteoarthritis of rat of blood stasis syndrome of inflammation therapy effect,for the clinical treatment of osteoarthritis and licorice aconite decoction and clinical application of huangqi guizhi wuwu tang to provide experimental basis.Methods:In Experiment 1,60 ICR mice were screened for pain threshold and grouped to prepare cold-dampness syndrome model.After 14 days of administration,the hot plate experiment was conducted.The pain threshold time of mice was observed and recorded at 45,90,135 and 180 min after the last intragastric administration.The writhing test was carried out in 0.6% glacial acetic acid solution(0.1 m L/10 g)after the same method model was replicated and the writhing times of mice were detected.In experiment 2,70 SPF female SD rats were prepared with cold coagulation and blood stasis knee osteoarthritis model by cold fixation method.The overall condition of rats in each group was observed and recorded.Arthritis index was assessed,body weight,foot palm temperature and knee diameter were measured,and the degree of joint swelling was calculated.The pathological changes of knee bone and joint were observed by HE staining.Serum IL-1β,IL-6 and TNF-α were detected by ELISA.PCR detection NF-B p65,TNF-α,IL-6m RNA relative expression in synovium and cartilage.Results:Experiment 1: The analgesic effect of Radix Glycyrrhiza Aconite Decoction and Huangqi Guizhi Wuwu Decoction on mouse pain model In the hot plate test,compared with the normal group,the pain threshold time of mice in the cold and dampness syndrome model group was longer.Compared with the model group of cold and dampness syndrome,the licking time of mice in treatment groups was longer after administration,and the licking time of mice in the combined prescription group of cold and dampness syndrome was longer at 135 min and 180 min after administration(P<0.05),the licking time of celecoxib group with cold and dampness syndrome at 180 min was longer(P<0.05).In the writhing experiment,compared with the model group with cold and dampness syndrome,the writhing times of the combined square group with cold and dampness syndrome decreased(P<0.05),and the incubation time was prolonged(P<0.05),the number of writhing was decreased and the latency time was prolonged in cold-dampness celecoxib group(P<0.05);Compared with the combined formula group,the number of body twisting in the combined formula group with cold and dampness syndrome decreased(P<0.05).Experiment 2: Experimental study of glycyrrhiza aconite decoction combined with huangqi guizhi wuwu decoction on rats with Yang deficiency and cold congealment osteoarthritis1.During model replication,the body weight of rats in each group decreased significantly,stool gradually became thin and soft,hair color became dull and lost luster,did not like exercise,and lower limb joints were swollen;After administration,compared with model group,the weight gain trend of Glycyrrhiza aconitum decoction group,Hefang group and celecoxib group became significantly higher,the hair color gradually recovered luster,the lower extremity activity was strong,the swelling of knee joint and toe palm of lower extremity gradually subsided significantly,and the amount of diet and drinking water gradually increased.2.Compared with the normal group,the arthritis index of rats in the model replication group increased during the model replication period,and reached the highest at the sixth week of model replication,the difference was statistically significant(P<0.05),at week 4 of administration,the arthritis index of each administration group was significantly decreased,compared with the model group,the difference was statistically significant(P<0.05).3.Pathological sections of the knee joint showed that the cartilage on the surface of the knee joint of rats in the model group showed defects,the thickness of the chondrocyte layer was thinned,the number of bone cells was significantly reduced,and the synovial cells were irregular.The surface of bone and joint of rats in the combined formula group was relatively smooth,with a little inflammatory cell infiltration,and no inflammatory infiltration in synovial tissue.4.Compared with the normal group,the contents of inflammatory factors IL-1β,IL-6 and TNF-α in the model group were all increased(P<0.05),compared with model group,the contents of inflammatory factors IL-1β,IL-6 and TNF-α in cecoxib group,Glycyrrhiza Fuzi decoction group,Hefang group and Huangqi Guizhi Wuwu decoction group were decreased(P<0.05).5.Compared with the normal group,the m RNA expression levels of NF-?B p65,TNF-α and IL-6 in synovium and cartilage of rat joints in model group were increased(P<0.05).Compared with model group,NF-?B p65 of cartilage tissue in celecoxib group,licorice aconite decoction group,Hefang group,celecoxib group,Huangqi Guizhi Wuwu decoction group and Hefang group had statistically significant differences in the relative expression of IL-6 in cartilage tissue(p<0.01).Conclusion:(1)based on haemorrheological nature osteoarthritis pathogenesis of blood stasis type,select the synopsis of licorice soup with huangqi guizhi wuwu decoction and lateral root of aconite,severely,homology of qi and blood,spleen and kidney,should both functions of dehumidification,warm through the cold,the origin of the chill,in addition to table and tonifying qi and blood,spleen and replenishing qi,to fill in the qi and blood deficiency syndrome,which can achieve the purpose of the cure for osteoarthritis.(2)For mice with cold and dampness syndrome and pain model,licorice aconite decoction and huangqi guizhi Wuwu decoction have the effect of nourishing blood,warming meridians and clearing colllaterals,dispersing cold,dehumidifying and relieving pain,dispersing cold and dampness in the body,nourishing blood and clearing colllaterals,thus improving the pain field value of mice with hot plate model of cold and dampness syndrome and twisting body model of cold and dampness syndrome.(3)Glycyrhiza aconite decoction and Huangqi Guizhi Wuwu decoction can inhibit the expression of inflammatory factors IL-1β,IL-6,NF-P65 and TNF-α in the NF-?B signaling pathway,and inflammatory factors such as IL-1β,IL-6 and TNF-αcan feedback regulate the NF-proliferation B signaling pathway and reduce the inflammatory response.(4)It is speculated that the analgesic effect of licorice aconite decoction and Huangqi Guizhi Wuwu decoction on rats may be related to the reduction of inflammatory response.The experimental study on the effectiveness of licorice aconite decoction and Huangqi Guizhi Wuwu decoction on the experimental model of pain in mice also showed that the effect of traditional Chinese medicine prescriptions was better for the animal model combining disease and syndrome.From the perspective of traditional Chinese medicine theory,aiming at the pathogenesis of cold coagulation and blood stasis in osteoarthritis,licorice aconite decoction and Huangqi Guizhi Wuwu decoction can relieve symptoms related to cold coagulation and blood stasis type osteoarthritis.From the experimental study,it can reduce the inflammatory reaction of osteoarthropathy and achieve the purpose of analgesia.
