Primary Pharmacodynamics Research On Jiawei Huangqi Guizhi Wuwu Decoction In Treating Experimental Rheumatoid Arthritis

Objective: Through establishing the autoimmune arthritis model, non-specific inflammation model,and the pain model, this article studies the therapeutic effect of the Jiaweihuangqiguizhi decoction(JWHQGZ) on the experimental rheumatoid arthritis, preliminary discusses its mechanism,in order to provide pharmacodynamic experimental basis for clinical application.Methods:Through injecting the Freund’s complete adjuvant into rats toes, inducedadjuvant arthritis(AA) model,that is the model of the immune arthritis,then, Different doses of JWHQGZ decoction and Prednisone Acetate were administrated on therat model. Observing the swelling of the joints,arthritis index(AI),weight of rats and the content of IL-1β、TNF-α in blood,find the mechanism of action of JWHQGZ.Furthermore,observed the effects of analgesic and non-specific anti-inflammatory by taking the experiments of hot plate test, writhing test, ear swelling test, acute paw swelling test.Results:1.The effects of JWHQGZ on immunologic inflammation of AA ratsThe high doses of JWHQGZ had the obvious inhibiting effect to the primary f-oot swelling of AA rats.Comparing the normal group,the rats of model group haveswelled obviously on toes after injecting18h and72h,especially after18h.The differences between high doses and low doses of JWHQGZ on swelling of the jointsare significant(P<0.05).Otherwise, the high doses of JWHQGZ have the obviousi-nhibiting effect to Secondary foot swelling.Comparing with the model group, thehigh doses group have the inhibiting effect after26d,which are made inflammatory(one day after the last administration),and the differences is obviously(P<0.05).Theswell of low doses of JWHQGZ is less than the model group,and no significant(P>0.05).The body weights of the high doses group of JWHQGZ are higher than the m odel group after making inflammation19d(gived drugs13d),23d(gived drugs17d),26d,and the differences is obviously. The arthritis index(AI) of the high doses group of JWHQGZ is declining,after making inflammation26d.At the same time,comparing with the model group,the differences is significant(P<0.05),and the low dos-es group is no significant(P>0.05).After making inflammation19d,the content of TNF-α in bloodof the high dosesgroup are declining obviously(P<0.01),but there is no significanton IL-1β(P>0.05).After making inflammation26d,TNF-α and IL-1βare both declined obviously. At t-he same time,the low doses group only decline the content of IL-1β at makinginflammation26d,it is nosignificant about the content of TNF-α(P>0.05).Above all,after making inflammation26d,the high doses group can decline the content of IL-1β and TNF-α clearly.2.the observation of the effects of analgesiaCompared with the previous administration,after gived drugs90min,the high dos-es group and low doses group of JWHQGZ both can raise the pain thresholdclearly(P<0.01);the high doses group can last almost120min after gived drugs(P<0.01).90min、120min pain threshold and increase the rate of the control group,the differencewas statistically significant(P<0.01)after the administration of high dose group.All groups which are gived drugs can reduce the times of body sprain causedby acetic acid(P<0.01),and prolonged latent period of twisting(P<0.01or P<0.05),itcan significant show the effects of analgesia maked by acetic acid.3.the observation of the effects of anti-inflammatoryComparing with normal groups,the high doses groups can reduced toes swellingcaused by carrageenin(P<0.01),but the low doses group only reduce the toes swelling after making inflammation4h(P<0.05).All groups which are gived drugs can reduce the ear swelling caused by dimet-hyl benzene clearly(P<0.01or P<0.05).Eeach dose group xylene-induced ear ede-mainhibition in mice was weaker than in the aspirin group.Conclusion:JWHQGZ has significant preventive and therapeutic effects on AA rats.One of the mechanisms maybe through adjust the factors such as IL-1β and TNF-α.Further more it has obviously the effect of anti-inflammatory and analgesia.
