Objective:Discussing the correlation between endoplasmic reticulum stress and diabetic peripheral neuropathy in ying wei qi xue insufficiency through analyzing and sorting the pattern and treatment theory of blood impediment of Zhongjing.Using MKR mice as experiment subject,discussing the mechanism of Huangqi Guizhi Wuwu decoction improve diabetic peripheral neuropathy through observing the affections of Huangqi Guizhi Wuwu decoction to glucolipid,metabolism and sciatic nerve of MKR mice.Using immunohistochemical techniques,WB,ELISA and other technical methods,on the one hand,we explored the relationship between endoplasmic reticulum stress and the onset of DPN under the pathogenesis of Yingwei Qi and blood deficiency;The effect of Guizhi Wuwu Decoction on DPN blood glucose,glucose tolerance,sciatic nerve conduction velocity,behavior,etc.,to explore the mechanism of Huangqi Guizhi Wuwu Decoction in improving peripheral neuropathy of type 2 diabetes.Methods:Theoretical research part.sorting the relevant articles in Zhang Zhongjing’s The Synopsis of the Golden Chamber,summarize the pathogenesis of hemorrhage,the characteristics of syndrome differentiation and treatment,and the physical characteristics of those who live in comfort,and analyze its value for modern understanding and treatment of diabetic peripheral neuropathy.Experimental research part.Sixty 8-week-old MKR mice were selected,males and females are half,fedding with high-fat diet for 4 weeks,and STZ solution was injected intraperitoneally to create a model.After the model of type 2 diabetes is formed,ice packs are given cold stimulation for 1 hour every day,and then the heat pain tail flick time,hot plate licking time and sciatic nerve conduction velocity are measured to determine the status of diabetic peripheral neuropathy.Exclude unmodeled mice.After feeding for 6 weeks,they are randomly divided into 5 groups:Group,Huangqi Guizhi Wuwu Decoction Group,Huangqi Guizhi Wuwu Decoction Group,Mecobalamin Group;10 MKR rats and FVB rats of the same age were set as the blank group.The mice were sacrificed 28 days after the administration.The fasting blood glucose of the mice was measured by electrochemical method,and the changes of blood lipids TCHOL and TG were measured by biochemical immunoluminescence method.The morphology of sciatic nerve tissue was observed by HE staining,Masson staining,and electron microscope.Western blot,Immunohisto-chemical detection of IRE1-α,p-JNK,JNK protein expression in sciatic nerve tissue.Result:Theoretical research shows that 1.Zhang Zhongjing’s theory of ying and wei believes that there are four pathological states of ying and wei: ying is insufficient and wei is sufficient,wei is insufficient and ying is sufficient,ying and wei are both sufficient,ying and wei are both insufficient.The same origin is derived from the metaplasia of the spleen and stomach,so the spleen and stomach should be treated when reconciling Yingwei.2.Zhang Zhongjing believes that the pathogenesis of blood impediment is the deficiency of Yingwei Qi and blood.The formation of Yingwei Qi and blood is closely related to the spleen and stomach,so the essence is the deficiency of the spleen and stomach.3.People with type 2 diabetes have a high degree of similarity in physical characteristics with those who live in comfort.Diabetic peripheral neuropathy and blood impediment also have a high degree of similarity in pathogenesis.The treatment is centered on the spleen and stomach,replenishing qi with warming the meridians and promoting blood flow with removing obstruction.The results of animal experiments showed that compared with the FVB group and the blank group,the general condition of the mice in the model group was worse,the fasting blood glucose was significantly increased,the serum TCHOL content was increased,the sciatic nerve function was decreased,the morphology and structure were disordered,and obvious pathological changes appeared,which proved DPN.The model is successful.Compared with the model group,the general condition of the mice in the administration group was improved,the mental state was better,the fasting blood glucose level decreased,the serum TCHOL content decreased(P<0.05),the serum TG decreased,the sciatic nerve function improved,and the morphological structure Relatively complete;the expression of IRE1-α and p-JNK protein in the original prescription group,Huangqi Guizhi Wuwu Decoction group and mecobalamin group were all decreased(P<0.05).Conclusion: 1.Through the discussion of Zhang Zhongjing’s theory of Yingwei,it is concluded that conditioning the spleen and stomachspleen is very important to reconcile Yingwei.2.Through the explanation of the pathogenesis of Blood impediment and the characteristics of those who live in comfort,it is concluded that Huangqi Guizhi Wuwu Decoction represented by the effect of replenishing qi with warming the meridians and promoting blood flow with removing obstruction are also important treatment methods for diabetic peripheral neuropathy.3.Huangqi Guizhi Wuwu Decoction plays a role in protecting sciatic nerve function and improving hyperalgesia by improving glucose and lipid metabolism,reducing endoplasmic reticulum stress and reducing endoplasmic reticulum stress damage.
