| Objective Study on the animal model of prethrombotic state in rats with early pregnancy and blood stasis by tonifying kidney and regulating blood and qi method,exploring the effect of Chinese medicine for invigorating kidney and regulating blood and preventing blood circulation on prevention of spontaneous abortion caused by prethrombotic state,and correlating from serum coagulation-anticoagulation-fibrinolysis.From the perspective of factors and coagulation function,this paper explores the possible mechanism of Chinese medicine for invigorating the kidney and regulating blood and preventing blood circulation to prevent spontaneous abortion caused by prethrombotic state.Methods Sixty SD pregnant rats were randomly divided into normal group(group A),model group(group B),low-dose group of traditional Chinese medicine(group C),medium-dose group(group D),and high-dose group of traditional Chinese medicine(group E),Aspirin group(group F).There were 10 in each group,the normal group was not modeled,and the other 5 groups were modeled with injection of epinephrine+mifepristone.All rats were given uniform gavage from1 d of pregnancy,once a day,and given for 11 days.Groups A and B were intragastrically administered with 10 m L/(kg · d)distilled water daily.Groups C,D,and E were intragastrically administrated with Chinese herbal medicine and qi and blood method to concentrate the decoction.The doses were 13.05g/(kg · d),26.1g/(kg · d),and52.2g/(kg · d).Group F was intragastrically administered with aspirin solution at a dose of 6.75mg/(kg·d).On the 11 th day of pregnancy,the rats were anesthetized with chloral hydrate by intraperitoneal anesthesia for 2 hours after the last administration.The abdominal cavity of the pregnant rats was dissected,and the number of embryo survival,the number of embryos lost and the embryo loss rate were calculated.Whole blood was taken from the abdominal aorta.APTT,PT,TT and FIB values were detected by automatic blood coagulation analyzer.Serum D-D,F1+2 and AT-? content were determined by ELISA.Results(1)The number of surviving embryos was 15.00±2.40 in the normal group and 4.800±1.99 in the model group;the model group was significantly lower than the normal group(P<0.05);the number of embryos lost was 9.10±1.85 in the model group;the normal group was0.80±0.92,the model group was significantly higher than the normal group(P<0.05);the embryo loss rate was 65.24±8.80% in the model group and 4.84±5.56% in the normal group,and the model group was significantly higher than the normal group(P<0.05);(2)Compared with the model group,the number of surviving embryos in the three different dose groups of Chinese medicine increased significantly,and the number of embryo loss and embryo loss rate decreased significantly(P < 0.05);compared with the aspirin group,the number of surviving embryos and embryo loss in the middle and high dose groups of traditional Chinese medicine The number and embryo loss rate improved significantly(P<0.05);the number of embryo loss and embryo loss in the low dose group were significantly lower than those in the aspirin group(P < 0.05);(3)After treatment,the plasma levels of APTT,PT and TT were shortened;compared with the model group,the middle dose group of Chinese medicine could prolong the APTT,PT and TT of the model rats(P<0.05),and the high dose group of Chinese medicine can prolong APTT,PT(P < 0.05),aspirin group can prolong TT(P <0.05);Compared with the model group,there was no significant difference in APTT,PT and TT between the low dose groups of traditional Chinese medicine(P > 0.05);the high-dose Chinese medicine group was superior to the aspirin and low-dose group of traditional Chinese medicine in prolonging the plasma APTT and PT values of rats with blood stasis(P < 0.05);(4)After the model establishment,the plasma levels of FIB,serum F1+2 and serum D-D were increased.The three doses of Chinese herbal medicine for invigorating the kidney and regulating blood and aspirin reduced plasma FIB,serum F1+2 and serum D-D levels in rats with blood stasis and abortion,which has a certain effect(P<0.05).Conclusions(1)A rat model of blood stasis induced by adrenaline epinephrine+mifepristone was successfully modeled in rats with premature thrombus in early pregnancy.(2)Chinese herbal medicine for invigorating the kidney and regulating qi and blood can reduce the embryo loss rate and the number of lost embryos in the model of blood stasis and abortion,increase the number of surviving embryos,and promote embryonic development,safe and effective;(3)The mechanism of Chinese medicine for invigorating the kidney and regulating qi and blood to improve the prethrombotic state of blood stasis model rats may be to prolong the PT,APTT and TT values of blood stasis model rats,reduce plasma FIB content,and reduce serum D-D and F1+2 content is achieved. |
