Objective Redox-disorder is a mediator of the disease occurrence and development,whose changes reflect the pathophysiological state of the lesion.We make efforts to explore the expression of(hydrogen polysulfide)in different gastric mucosa diseases by using fluorescent probes to detect an important reducing substance in the body,and to explore the inflammation,intestinal metaplasia,and malignancy of hydrogen polysulfide in the stomach.The characteristics of the change,and discuss its possibility as a biomarker.Methods With the help of "off-on" near-infrared fluorescent probe BD-ss,the hydrogen polysulfides of human gastric cancer cell line(HGC-27 cell line)and mice stomach,combined with pathological features,are measured by confocal laser scanning microscope or flow cytometry and small animal imaging system,respectively,to detect the fluorescence features in inflammation,intestinal metaplasia and malignancy.Results The fluorescent intensity of HGC-27 cells increased with the growth factor or inflammatory factor stimulation compared with that of untreated cells after being incubated by BD-ss(P<0.001).And oxaliplatin could reduce the increase of cellular fluorescence intensity under malignant proliferation.The stomach fluorescence intensity enhanced of the chronic inflammation mice(P<0.05)and the gastric cancer mice(P<0.01).In addition,the fluorescence in cancerous stomach retained longer than that of chronic inflammation stomach with the result of small animal imaging system.The flow cytometry results of endoscope samples showed that the fluorescence of chronic inflammation(P<0.01),intestinal metaplasia(P<0.05)and malignant tumors(P<0.01)increased compared with normal gastric mucosa.Especially,the fluorescence of malignant tumors is much higher than other two groups.Conclusion In the state of inflammation and hyperplasia of the stomach,polysulfide increases,and it is more obvious in the malignant proliferation state.It is related to the disease state of the stomach.The changes can reflect the disease state and can be used to diagnose the disease and provide quantitative data,as well as the results of the diagnosis.
