【Objective】To investigate the effects of hydrogen sulfide on the injury of colon mucosa of septic rats through the construction of the model of sepsis and provide new theoretical basis and treatment strategies for sepsis.【Methods】Male SD rats were divided into 5 groups: (1)control group (2) sepsis group (3) sepsis+NaHS group (4) sepsis+DL-propargylglycine (PAG) group (5) sepsis+ aminooxyacetic acid (AOAA) group. Rats were subjected to cecal ligation and puncture (CLP)- induced sepsis and treated with saline(ip),NaHS(10mg/kg, ip),PAG(50mg/kg, ip),AOAA(20mg/kg, ip) 30 minutes before CLP respectively. H2S synthesis, the concentration of TNF-α,IL-1β, the activity of MPO , cystathionine-γ-lyase (CSE) mRNA level and cystathionine-β-synthase (CBS) mRNA level in colon were determined 20 hours after CLP. The morphological structure and infiltration of inflammatory cells of colon tissue were also observed. 【Results】①When compared with normal group, the H2S synthesis, the concentration of TNF-α,IL-1β,the activity of MPO, CSE mRNA level and CBS mRNA level in colon of sepsis group were increased (P<0.01), the structure of colon was damaged and more inflammatory cells infiltrate into the tissue.②When compared with sepsis group, the H2S synthesis, the concentration of TNF-α,IL-1βand the activity of MPO were increased in sepsis+NaHS group(P<0.05), but a decrease of CSE mRNA level, and CBS mRNA level(P<0.01).The damage of colon structure was worse than sepsis group. On the other hand, the administration of AOAA could significantly reduce the systemic inflammation of sepsis, as evidenced by decrease of TNF-α,IL-1β, the activity of MPO(P<0.01)and less damage of colon structure. The H2S synthesis ,the concentration of TNF-α, IL-1βand the activity of MPO were also decreased in sepsis+PAG group, but with no statistically difference.【Conclusion】When sepsis occurs, more H2S was produced primarily by the catalysis of CBS in colon, play a proinflammatory role and aggravate colon mucosa injury.
