Objective:1.To verify the effect of acupuncture on cognitive memory in rats with vascular dementia(VD)by comparing the Morris water maze test results of each group of VD rats.2.To observe the effect of acupuncture on the morphology of hippocampus in rats with vascular dementia by HE staining.3.To verify the effect of acupuncture on Ngb and HIF-1? gene transcription and protein expression in hippocampus of rats with vascular dementia.Methods:1.The model of vascular dementia Rats was made by improving the internationally recognized method of 4-VO.2.Twelve rats were selected as the sham operation group,and expose the four blood vessels of the rats,without special treatment.The 36 successful rat models were randomly divided into model group,electroacupuncture group and drug group,each group of 12.The electroacupuncture group was given acupuncture Baihui and Fengfu.And at the same time,electroacupuncture stimulation of these two points.Stimulus current 1m A,frequency 2Hz.Treatment once a day,each time lasted 30 minutes,1 day off after 6 days of treatment,a total of 4 weeks treatment.The drug group was treated with Periplum Intragastric administration)and started treatment at the same time as the Electroacupuncture group.Treatment once a day,1 day off after 6 days of treatment,a total of 4 weeks treatment.The model group and the sham operation group only perform grasping and fixation without any treatment.3.Respectively do the Morris water maze test after modeling and after treatment.Screen out the successful rat models and detect the improvement of the learning and memory in rats.4.Observing the morphological changes of hippocampal neurons using HE staining.5.Qualitative and quantitative analysis of Ngb and HIF-1? proteins by Western blot(neuron: Neurofilament(NF),astrocytes: using glial fibrillary acidic protein(glial fibrillary acidic protein)GFAP).6.Detect the expression of Ngb,HIF-1? and p53 RFP m RNA by RT-PCR.Results:Compared with the sham operation group,the model group was significantly prolonged in the water maze test,and the number of crossing platforms was significantly reduced(p<0.05).After treatment,compared with the model group,the escaping latency was significantly shortened,and the number of crossing platforms increased significantly in electroacupuncture group and drug group(p<0.05).And the electroacupuncture group is more obvious than the drug group(p<0.05).The HE-stained neurons were observed under a 400-fold microscope.The number of neurons in the hippocampal CA1 area of the model group was significantly reduced,and some neurites were pathologically shortened or even disappeared.Compared with the model group,the number of nerve cells in the hippocampal CA1 area of the electroacupuncture group and the drug group increased,the arrangement was relatively neat,the cell structure and morphology were basically normal,the degree of cell edema was lighter,and the electroacupuncture group was more obvious.Compared with the model group,the electroacupuncture group and the drug group showed that the degree of brain neuron damage was significantly reduced,the expression of Ngb,HIF-1 ?,NF m RNA and protein was significantly increased,and the expression of p53 m RNA and GFAP m RNA and protein were significantly decreased(p<0.05).And the improvement of the electroacupuncture group was more significant than that of the drug group(p<0.05).Conclusion:Acupuncture "Baihui" and "Fengfu" can improve the water maze performance of rats with vascular dementia and improve their learning and memory ability.By promoting the expression of Ngb,HIF-1?,NF in the hippocampus of the brain and reducing the content of P53 and GFAP,it can inhibit the apoptosis of neuronal cells.
