Effects Of Nuclear Factor Expression In Hippocampal Nerve Cells Of Epileptic Rats Through Acupuncture Manipulation On Governor Vessel

Objective:To explore the possible mechanism of acupuncture in the treatment of epilepsy by acupuncture governor vessel points(Baihui,Fengfu,Dazhui,Taodao,Wuming and Shenzhu).Methods:The model of epilepsy was established by lithium chloride-pilocarpa method.The rats were randomly divided into three groups: EP(epilepsy)model group,western medicine group and acupuncture group,16 rats in each group.Another 16 rats in the same batch as the normal group.Once modeling success,the day began treatment.Acupuncture group was treated by acupuncture governor vessels(Baihui,Fengfu,Dazhui,Taodao,Wuming and Shenzhu).Western medicine group selected sodium valproate(300mg/kg)for treatment.The blank group and the EP model group were not treated.The course of treatment is 14 days,once a day.At the end of the treatment,Morris water maze test was performed.The expression of NF-?B in hippocampus was detected by immunohistochemical staining.The expression of NF-?B(P65)protein in hippocampus was determined by immunohistochemical staining.The expression of NF-?B mRNA in hippocampus was detected by PCR.Result:(1)General state and behavioral observationFour groups,the blank group of rats in general the best,and no seizures.EP model group of rats in general the worst,poor diet,weight loss,coat yellowing off,slow response,often accompanied by spontaneous recurrent epilepsy behavior.Acupuncture group and western medicine group were significantly improved compared with EP model group(P <0.05),and the duration of seizure duration and the frequency of seizure were lower than those of EP model group(P <0.05)The difference was not significant(P> 0.05).(2)Morris water maze test resultsThe mean escape latency and average swimming distance were shorter in the blank group than in the EP model group(P <0.05).The acupuncture group and the western medicine group were also shorter than those of the EP model group(P <0.05).There was no significant difference between the two groups(P <0.05),but there was no significantdifference between the two groups(P> 0.05).(3)HE observation under light microscopeThe results showed that the hippocampus cells in the blank group were well arranged and the cell morphology was normal.The hippocampal cells in the EP model group were loose,the cells were dissolved,deformed,chromosomes,And even disappeared;acupuncture group,Western medicine group hippocampus cells in different degrees of injury,but compared with the EP model group has improved.(4)Immunohistochemical staining of the identification resultsThe cytoplasm of hippocampal cells in the blank group was positive for NF-?B(P65),and the nuclei of the hippocampus were positive in the EP model group.The cytoplasm and nucleus of the acupuncture group and the western medicine group were positive.The results showed that the IOD value(mean optical density)of hippocampus NF-?B(P65)in hippocampus of EP model group was higher than that of blank group(P <0.05),and the IOD value of western medicine group and acupuncture group decreased(P < 0.05),the difference was statistically significant.There was no significant difference between acupuncture group and western medicine group(P> 0.05).(5)PCR was used to detect the expression of NF-?BThe NF-?B mRNA in hippocampus of EP model group was significantly higher than that of normal group(P <0.05).The expression of EP model group was significantly higher than that of EP group(P <0.05),and the difference was statistically significant(P <0.05),acupuncture group,There was no significant difference between the two groups(P> 0.05).Conclusion:The acupoints of acupuncture points can improve the learning and memory ability and growth status of EP rats and reduce the time and frequency of seizures in EP rats.The mechanism may be related to the inhibition of NF-?B mRNA and protein expression in hippocampus.
