| Objective: The expression of Ras converting enzyme 1(Rce1)in gastric cancer tissues was detected by IHC-P and Western blot method.To explore the relationship between Rce1 and the occurrence,development and prognosis of gastric cancer,and to provide a basis for the decision making of comprehensive treatment for gastric cancer.Methods: From December 2017 to December 2018,12 patients who were diagnosed with gastric cancer by gastroscopy and underwent radical gastrectomy in Sichuan mianyang 404 hospital(The second affiliated hospital of North Sichuan Medical College)were collected.Gastric cancer tissues were cut out during the operation as experimental group 1(Western blot,n=12).Meanwhile,70 paraffin specimens of gastric cancer undergoing radical gastrectomy in Sichuan mianyang 404 hospital province from December 2014 to December 2015 were collected as experimental group 2 together with the above 12 gastric cancer specimens(IHC-P,n=82).A total of 12 patients who underwent subtotal gastrectomy for massive gastric ulcer or perforation of gastric ulcer in Sichuan mianyang 404 hospital province from December 2017 to December 2018 were collected.Benign gastric lesion tissues were taken during the operation as control group 1(Western blot,n=12).Collected in December 2017-December 2018 in Sichuan mianyang 404 hospital outpatient gastroscope pathological examination diagnosis of 20 cases of gastric ulcer patients,gastroscope collect gastric benign lesions tissue samples(n = 20),and 12 cases with huge gastric ulcer or gastric ulcer perforation most of gastric resection of gastric benign lesions tissue samples(n = 12)of the control group 2(IHC-P,n = 32).Comparative analysis Rce1 expression in gastric cancer and gastric benign lesions tissue level difference,analyzing Rce1 expression level and gastric cancer patients' age,gender,tumor stage,the degree of differentiation,histological type,tumor size,lymph node metastasis,the relationship between the collected at the same time corresponding to the survival of patients with gastric cancer and 70 cases of paraffin specimen data,analysis Rce1 expression level relationship with stomach cancer patients survival time.Results: Western blot detection of Rce1 protein expression in gastric cancer tissues and benign gastric lesions: Western blot detection found that the expression level of Rce1 protein in 12 gastric cancer tissues was higher than that in 12 benign gastric lesions[(0.46 0.32)vs.(0.15 0.27),t=2.582,P=0.017].IHC-P detection Rce1 expression in gastric benign lesions organizations: 20 cases of outpatient gastroscope gastric benign lesion group and 12 cases because of the huge gastric ulcer or gastric perforation line just 7 cases of surgical treatment for gastric benign lesion tissue Rce1 went into a state of high expression,high expression rate is only 21.88%(7/32),there are 25 cases Rce1 present a lower expression condition,the low expression rate was 78.12%(25/32).Expression of Rce1 in gastric cancer tissues detected by IHC-P: among 82 gastric cancer tissues,61 showed high expression of Rce1,with a high expression rate of 74.39%(61/82),and 21 gastric cancer tissues with low expression of Rce1,with a low expression rate of 25.61%(21/82).The expression level of Rce1 in gastric cancer tissues was higher than that in benign gastric lesions(P < 0.05),which was consistent with Western blot results.The study found that in different TNM staging of gastric cancer tissues Rce1 expression level is different,Rce1 in phase I,II,III gastric cancer tissue high expression rate is respectively: 47.06%(8/17),76.74%(33/43),90.91%(20/22),the phase III and phase II Rce1 expression in gastric cancer tissue level obviously higher than that of stage I cancer of the stomach tissue(P < 0.05),Rce1 expression level in stage III gastric cancer tissues and no statistical difference between stage II gastric cancer tissue(P > 0.05);High expression rate of Rce1 in tumor tissues of patients with tumor diameter 4cm and tumor diameter < 4cm [87.88%(29/32)vs.65.31%(32/49),x2=5.274,P=0.022].High expression level of Rce1 in tumor tissues with and without lymph node metastasis [81.87%(49/60)vs.54.55%(12/22),x2=6.215,P=0.013].Differentiation of low,medium and high differentiation of gastric cancer tissues Rce1 high expression level are: 91.43%(32/35),65.38%(17/26),57.14%(12/21),low differentiation of gastric cancer tissues Rce1 expression level was significantly higher,high differentiation in gastric cancer tissue(P < 0.05),the differentiation of gastric cancer tissues Rce1 expression level and high differentiation of gastric cancer tissues Rce1 expression level no statistical difference(P > 0.05).There was no significant relationship between the expression level of Rce1 and patients' gender,age and histological type(all P > 0.05).Follow-up data of 70 patients with gastric cancer: 1 patient in the low expression group of Rce1 was lost to follow-up at 21 months after surgery,and 2 patients in the high expression group of Rce1 were lost to follow-up at 18 months and 21 months after surgery,respectively.The 1-year,2-year and 3-year survival rates of gastric cancer patients in the low expression group of Rce1 were 94.12%,81.25% and 75.00%,significantly higher than those in the high expression group of Rce1(88.68%,50.98% and 45.10%),with statistically significant differences(P < 0.05).Conclusion:(1)The expression level of Rce1 in gastric cancer tissues was significantly higher than that in benign gastric lesions.(2)The expression level of Rce1 was correlated with the clinicopathological characteristics of gastric cancer,such as TNM stage,tumor size and tumor differentiation degree.With the development of TNM staging,tumor enlargement and differentiation of gastric cancer,the expression level of Rce1 was higher.(3)The expression level of Rce1 is negatively correlated with the 3-year survival rate of gastric cancer patients,and the expression level of Rce1 can be used as a reference for the prognosis evaluation of gastric cancer patients after surgery. |
