Objective: To investigate the effect of p EGFP-N1-IL-17 on the apoptosis of Hep-2cells and to explain the mechanism by which p EGFP-N1-IL-17 inhibits the apoptosis of Hep-2 cells through the Fas/Fas L signaling pathway.Method: The recombinant plasmid vector p EGFP-N1-IL-17 carrying IL-17 gene was constructed and transfected into Hep-2 cells,Normal Hep-2 cells were used as the control group,and p EGFP-N1-NC was used as negative control group.p EGFP-N1-IL-17 was used as the experimental group.The effect of p EGFP-N1-IL-17 on the apoptosis rate of Hep-2 cells was detected by flow cytometry.Western blot was used to detect the expression of Fas and Fas L in Hep-2cells transfected by pEGFP-N1-IL-17.Results: Annexin V-FITC /PI cell apoptosis assay results showed that the apoptosis rate of Hep-2 cells in experimental groups decreased obviously.The results of Western blot showed that the expressions of Fas,Fas L,Cleaved-Caspase8 and Cleaved-Caspase3 in experinental group of Hep-2 cells were significantly decreased(P<0.01).Conclusion: p EGFP-N1-IL-17 may inhibit the apoptosis of Hep-2 cells through FAS/FASL signaling pathway.
