| ObjectiveAim to observe the effect of curcumin,resveratrol and its combination on the proliferation and apoptosis of laryngeal cancer cells in vitro,and to investigate the possible mechanism of anti tumor action of Hep-2.MethodsCell growth inhibition rate was detected by MTT method;The morphological changes of the nucleus by Hoechst 33258 staining method;Apoptosis rate was detected by Flow cytometry;The expression of p65 and Caspase-3 gene was detected by RT-PCR;Blot Western method to detect the effects of the drug on the expression of p65 protein and Caspase-3 protein in laryngeal carcinoma cells.ResultsMTT results showed that the effect of curcumin combined with resveratrol on the proliferation of laryngeal cancer cells was significantly higher than that of curcumin alone and resveratrol alone(P<0.05);The results of Hoechst 33258 staining showed that the morphology of nuclei in each treatment group showed a characteristic change of apoptosis compared with the control group;The results of flow cytometry analysis showed that the apoptosis rate of curcumin combined with resveratrol group was significantly higher than that of curcumin and resveratrol alone group(P<0.05);RT-PCR results showed that the expression of p65 gene combined with resveratrol and curcumin group was two when used alone were significantly decreased(P<0.05),and Caspase-3 gene combined with resveratrol group curcumin expression and two when used alone were significantly higher than(P<0.05);Western Blot results showed thatcompared with the control group,the expression of p65 protein and curcumin group alone alone resveratrol group significantly decreased the expression of Caspase-3 protein were significantly up-regulated,while curcumin and resveratrol groups of p65 and Caspase-3 expression compared with single effect of curcumin and resveratrol alone group had significant difference(P<0.05).ConclusionCurcumin and resveratrol has synergistic effect on inducing apoptosis of human laryngeal carcinoma cell Hep-2,the mechanism is probably by blocking transcription factor kappa B NF-(p65)signal pathway activation,the up-regulated expression of Caspase-3 protein to achieve. |
