Objective To investigate the effect of Interleukin-17(IL-17)on the apoptosis of laryngeal squamous cell carcinoma and to elucidate the mechanism by which IL-17 inhibits the apoptosis of laryngeal squamous cell carcinoma by inhibiting the Fas / Fas L signaling pathway through the PI3 K / AKT signaling pathway.Method 1.Tumor specimens from Sixty patients who were pathologically diagnosed with laryngeal squamous cell carcinoma from March 2010 to December 2016 from The Second Hospital of Anhui Medical University were selected to take part in this study.According to the pathological grading,the tissue samples were divided into adjacent non-cancerous group,well differentiated group,moderately differentiated group,poorly differentiated group.The expression of IL-17,IL-17 R,PI3K,p-PI3 K,AKT,pAKT,Fas and Fas L in each group were detected by Western blot.2.We used q RT-PCR to detect the expression of IL-17,IL-17 R,Fas,Fas L m RNA in laryngeal cancer tissues and adjacent tissues.3.Western blot was used to detect the expression of PI3 K,p-PI3 K,AKT,p-AKT,Fas and Fas L in Hep-2 cells treated with different concentrations of IL-17.4.The expression of Fas/Fas L in Hep-2 cells stimulated with IL-17 following pre-treatment with PI3 K inhibitor.5.Flow cytometry was used to detect the effect of IL-17 stimulation on Hep-2 cell apoptosis.Result 1.The expressions of IL-17,IL-17 R,p-PI3 K and p-AKT in laryngeal squamous cell carcinoma tissues were significantly higher than those in paracancerous tissues(P <0.01;P <0.01;P < 0.01;P <0.01),the expression of Fas and Fas L were decreased in carcinoma tissues(P <0.01;P <0.01).With the increase of malignant degree of laryngeal carcinoma,the expression of IL-17 and IL-17 R increased gradually,and the expression of Fas and Fas L decreased gradually.2.The expression of IL-17 mRNA was negatively correlated with the expression of Fas and Fas L m RNA.The higher the expression of IL-17,the lower the expression of Fas and Fas L(r1 = 0.820,r2 = 0.8470 P <0.01).3.The results of Western blot showed that the expressions of p-PI3 K and p-AKT protein in Hep-2 cells were significantly increased with the increase of IL-17 concentration(P <0.01;P <0.01).Compared with the control group,the expressions of Fas and Fas L proteins in 1ng / m L group,50 ng / m L group and 100 ng / m L group were significantly decreased(P <0.01;P < 0.01;P < 0.01).5.The apoptosis rate of Hep-2 cells treated with 50 ng / m L IL-17 for 6h,12 h and 24 h were lower than in the control group.6.The expression of p-AKT in Hep-2 cells stimulated with IL-17 following pre-treatment with Wortmannin was significantly decreased(P <0.01;P <0.01),while Fas and Fas L were significantly increased(P <0.01).Conclusion: IL-17 can inhibit laryngeal cancer apoptosis through the PI3 K / AKT / Fas / Fas L signaling pathway.
