Basic Research And Pharmacological Analysis Of Turmeric Processed Coptidis Rhizoma Standard Pieces

Coptidis is the dried rhizome of Coptis chinensis Franch.,Coptis deltoidea C.Y.Cheng et Hsiao or Coptis teeta Wall.The Chinese Pharmacopoeia 2015 edition contains a copy of Coptidis Rhizoma,turmeric processed Coptidis Rhizoma and Euodiae Fructus-Coptidis Rhizoma.The quality standard of turmeric processed Coptidis Rhizoma is the same as that of Coptidis Rhizoma,lacking the special quality standard and standard substance(standard pieces).The quality of the pieces is difficult to control,which seriously affects the clinical efficacy and medication safety.The Coptidis Rhizoma is more bitter and cold,and the ginger juce processing can inhibit its bitter cold.However,the mechanism of Coptidis Rhizoma changes before and after turmeric is still unclear.In this paper,by preparating of turmeric processed Coptidis Rhizoma Standard Pieces and studying of quality standards and fingerprints,in order to standardize the preparation process of turmeric processed Coptidis Rhizoma Standard Pieces,improve the quality control standards,establish the special quality standards for turmeric processed Coptidis Rhizoma and the preparation of " standard pieces " lay the foundation.A preliminary study on the changes of medicinal properties before and after processing of turmeric was carried out in order to explain its processing mechanism and provide scientific basis for the clinical application of turmeric.1.Study on preparation technology of turmeric processed Coptidis Rhizoma Standard PiecesThe technical procedures of turmeric processed Coptidis Rhizoma,softening,cutting,drying and ginger processed were standardized.The technical parameters were determined and batch preparation was carried out.The turmeric processed Coptidis Rhizoma Standard Pieces of different granulation methods were used to pulverize and prepare different sizes of powder.And the composition content analysis and comparison,determined the method of homogenization of turmeric processed Coptidis Rhizoma Standard Pieces.2.Study on quality standard of turmeric processed Coptidis Rhizoma Standard Pieces According to the "Chinese Pharmacopoeia" of the 2015 edition of the " Coptidis Rhizoma " item,the traits,thin layer identification,moisture,total ash,leachables,content determination and other items of turmeric processed Coptidis Rhizoma Standard Pieces were studied.A thin layer identification method for the determination of gingerol in turmeric processed Coptidis Rhizoma was established.A method for determination of berberine,arbutin,jatrorrhizine,berberine,coptisine and palmatine was established by a multi-evaluation method.The quality standard of turmeric processed Coptidis Rhizoma Standard Pieces was formulated.3.Study on Fingerprint of turmeric processed Coptidis Rhizoma Standard PiecesThe HPLC method was established the fingerprint of the standard pieces of turmeric processed Coptidis Rhizoma,and 18 common peaks were indicated.The cluster analysis and Principal component analysis were used to analyze the specificity,which can effectively evaluate the quality of turmeric processed Coptidis Rhizoma.The comparison with the fingerprints of Coptis Rhizoma and Euodiae Fructus-Coptidis Rhizoma was different.4.Influencing factors and accelerated stability of turmeric processed Coptidis Rhizoma Standard PiecesInfluencing factors of turmeric processed Coptidis Rhizoma Standard Pieces with different particle sizes were investigated.After 10 days of high humidity,high humidity and light conditions,the water content and main active ingredients changed significantly,but the difference between different particle sizes was not significant.Accelerated stability test results under packaging(brown glass bottles,aluminum foil bags)showed that the content of active ingredients decreased with time,but the test indicators met the requirements;the difference of active ingredients content under different packaging conditions was not obvious.5.Study on the changes of pharmacological before and after processing of turmeric processed Coptidis Rhizoma Standard PiecesUsing pharmacological methods,research the hexokinase,fructose phosphokinase,pyruvate kinase,citrate synthase,and isocitrate dehydrogenation source of three key energy metabolism pathways,were administered to rats after oral administration of Rhizoma Coptidis and turmeric.The content and activity of enzyme,α-ketoglutarate dehydrogenase and respiratory chain complexes I,II,III and IV were determined.The results showed that both Rhizoma Coptidis and turmeric could significantly reduce the activity of the above enzymes,thereby inhibiting the body energy.Metabolism,but the inhibition of turmeric was significantly weaker than that of Rhizoma Coptidis.The main component analysis was used to analyze the content of key enzymes.The results showed that the effects of Rhizoma Coptidis and Curcumin on energy metabolism were mainly in the oxidative phosphorylation pathway,of which mitochondrial respiratory chain Complexes II and III are the main indicators of their differences.The medicinal changes of Phyllostachys pubescens and its relationship with energy metabolism were preliminarily elucidated.
