Objective Breast cancer is currently one of the most common female malignancies,and the global incidence is increasing year by year.Chinese medicine believes that breast cancer patients generally have qi and blood deficiency,so breast cancer should be treated through supplementing qi and blood.However,it's clinical efficacy on malignant tumors is still controversial.Therefore,we try to analyse effects of Danggui and Huangqi on tumor and peripheral blood of 4T1 breast cancer mice This experiment is to explore the effects of supplement qi and blood method on serum and tumor tissue IL-10 and TGF-?1 in 4T1 breast cancer mice by animal experiments,that give the treatment of Danggui and Danggui in 4T1 breast cancer mice under the guidance of the basic theory of Chinese medicine.Methods 72 female BALB/c mice were randomly divided into 9 groups: Normal group(N),Model group(M),Danggui-low dose group(DL),Danggui-medium dose group(DM),Danggui-high dose group(DH),Huangqi-low dose group(HL),Huangqimedium dose group(HM),Huangqi-high dose group(HH),Cyclophosphamide group(CTX).Respectively: N(0.9% Physiological saline 0.2m L every mince),M(0.9% Physiological saline 0.2m L every mince),DH(100mg/kg),DM(30mg/kg),DL(10mg/kg),HH(300mg/kg),HM(100mg/kg),HL(30mg/kg),CTX(30mg/kg).The orthotopic tumor,thymus,lung and spleen were dissected,weighed and calculated for each organ index;the texture and size of the organ were visually observed and photographed;the lungs were fixed with 10% formalin for 48 hours,visually observed and recorded.The number of lung metastases in each group;after formalin fixation of thymus,lung,spleen and tumor tissues,hematoxylin-eosin(H&E)staining was performed to observe histomorphology and pathological changes;immunohistochemistry(IHC)method,analysis the effect of Danggui and Huangqi on the expression of CD3 in thymus tissues and the effect of Danggui and Huangqi on the expression of IL-10 and TGF-beta1 in tumor tissues;blood routine test was used to compare the number of peripheral blood cells and the abnormality of the three-line structure in each group of mice;blood smear was used to observe the number of white blood cells in the peripheral blood and its nuclear lobes.The effects of Danggui and Huangqi on serum IL-10 and TGF-beta1 levels were analyzed by ELISA.Results In terms of blood,a certain dose of danggui can significantly increase the number and percentage of peripheral blood lymphocytes in 4T1 breast cancer mice,and a certain dose of Huangqi can significantly reduce the number and percentage of basophils.A certain dose of Danggui and Huangqi can significantly increase thymus weight and index.In terms of related organs,Danggui can significantly increase lymph nodes,lung weight and index.In terms of lung metastasis,both Danggui and Huangqi can reduce the number of large lung metastases,and Huangqi can also reduce the number of total lung metastases and lung metastasis.There were different degrees of improvement in lymphocytes in the thymus tissue of Danggui and Huangqi.Huangqi can significantly reduce IL-10 levels in serum of breast cancer mice.Danggui and Huangqi can significantly reduce the level of TGF-?1 in the serum of breast cancer mice.Danggui and Huangqi can significantly reduce the positive expression rate of IL-10 in tumors.Both Danggui and Huangqi can significantly reduce the positive expression rate of TGF-?1.Conclusion 1.Danggui and Huangqi can improve peripheral blood lymphocytes and basophils,improve thymus structure,increase thymus weight and index,reduce lung metastasis in 4T1 breast cancer mice and have certain anti-breast cancer effects.2.IL-10 and TGF-?1 may be one of the mechanisms of Danggui and Huangqi in improving peripheral blood and reducing lung metastasis in 4T1 breast cancer mice.
