Objective To observed the effect of modified huangqi danggui decoction effective components compatibility on MCAO rats nerve protection.Methods Extract Astragalus polysaccharides crude products, Angelica sinensis polysaccharide crude products and Hirudin crude products by Water-extraction and alcohol-precipitation method.Water liquid extraction to get Astragalus saponin crude products and heating reflux method to get Ferulic acid crude products.Line-plugged method to establish the rat middle cerebral artery occlusion(MCAO) model.MCAO rats were randomly divided into sham-operated group,model group,effective components group,and water decoction group.Water decoction group and effective components group were lavaged with huangqi danggui water frying concentrate and components compatibility mixed suspension with the volume-and-mass proportion of 1ml/100 g.Meanwhile, sham-operated group and model group were lavaged with equivalent physiological saline once a day,continuously for 14 days.Neuroethology was evaluated by Bederson grading, crossbar experiment and screen. Morphology change of hippocampal CA1 area was explored by HE staining and Nissl's staining.Neural cell apoptosis was detected with TUNEL method.Protein expression of PI3 K,Bcl-2,caspase-3 was detected with immunohistochemical method.VEGF and VEGFR2 mRNA protein in hippocampal CA1 area expression were detected with the RT-qPCR method and Western blotting method.Result(1) Astragalus polysaccharides crude products,Angelica sinensis polysaccharide crude products,Hirudin crude products, Astragalus saponin crude products and Ferulic acid crude products were extracted and approved.The content of Astragalus polysaccharide in terms of glucose was 37.89%,Angelica sinensis polysaccharide in terms of glucose was 24.69%, Hirudin 20U/g,Astragalus saponin content in astragalus glycosides calculation was 0.088% and Ferulic acid 0.050%.(2)Compared with sham-operated group,the Bederson score and screen test score of model group rats increased significantly,while crossbar experiment score decreased significantly(P<0.05);Compared with model group,Bederson score and screen test score of each dose group rats reduce significantly, crossbar experiment score rised significantly(P<0.05);(3)HE staining results shows a reduced numbers of rat hippocampus CA1 area neurons,and a sparse, disorder, widened-gap,partial necrosis around the cell;Compared with model group,the drug delivery in the rat hippocampus CA1 reduce pathological changes in different degree and the neuron loss decreased, cell arrangement was neat,and gap around the cell was narrowed.Nissl's staining results showed a severely-damaged, reducing number or disappeared of nissl bodies neurons in rat hippocampal CA1 area and Surface mottle;Compared with model group,neurons injury loss hippocampus CA1 area had witnessed a reduction in each dose group, cytoplasm nissl bodies of depigmentation can alleviate,shading is relatively uniform.(4)Compared wih sham-operated group,model group in the rat hippocampus CA1 area neurons apoptosis rate increased significantly(p<0.05);Compared with sham-operated group,each dose group rat hippocampus CA1 area neurons apoptosis rate was significantly lower(p<0.05);Compared with sham-operated group,model group rats hippocampus CA1 area was higher VEGF,VEGFR2 mRNA and protein expression, Caspase-9 protein expression significantly increased,Bcl-2 prorein expression significantly reduced(p<0.05);Compared with model group,theVEGF,VEGFR2 mRNA and protein expression in drug group rats hippocampal CA1 area increased,Caspase-9 protein expression decreased significantly,Bcl-2 protein expression was significantly higher(p<0.05).Conclusions Modified huangqi danggui decoction and components compatibility could improve the function of cerebral ischemic pathological damage degree,the hippocampus nerve protective effect,its mechanism may be related to the regulation of VEGF/VEGFR2 signal pathway,inhibition of cerebral ischemia hypoxia caused by neuron apoptosis related.The research results showed the scientific nature and rationality of effective components compatibility based on prescription compatibility and pharmacological mechanism.
