Objectives:The inflammation and immune systems to activate play an important role in As,also many immunity cells participate,include the monocytes,macrophages,lymphocytes and labrocytes etc.These cells produce a great deal of cytokines,such as the IL-1,TNF,TGF, MCP-1,ICAM-1 etc.They pass to lie to lead an immune response of cell,promote the inflammation but rise the important function in the As.Our research studied DuiGui BuXue Decoction and the combination of fractions extracted from HuangQi and DangGui effect on the TNF-α,ICAM-1's expression induced by Ox-LDL in RAW264.7cells,in order to discuss DuiGui BuXue Decoction stopped the pathological changes in early As.Methods:1.Extract and separate the fractions of HuangQi and DangGui.2.Prepared rabbit serum containing DuiGui BuXue Decoction herbs or not and rabbit serum containing the valid target of HuangQi and DangGui or not,Ox-LDL,cell activity by trypanblue staining method,detected cell survival rate affected by rabbit serum through MTT method.3.Detected TNF-α's protein activity change by WB method.4.Detected ICAM-1's protein activity change by WB method.Results:1.cells grew adherence to the well after 3 to 4 hours' culture,2 to 3 days later,cells grew fully monolayer,cell activity detected by trypan blue staining was more than 95%.Rabbit serum had nutrient action to facilitate cells' growth,but too highly concentration was bad to cells.10%concentration had small effect on cells. 2.TNF-α's WB results:Ox-LDL group protein density area ratio is 0.8997±0.0121, compared with control group(0.2033±0.0193),the difference had great meaning (p<0.01),implied after Ox-LDL stimulate RAW264.7 cells,TNF-α's protein activity obviously.Inhibitor group(0.3464±0.0203)compared with Ox-LDL group,difference was obvious(p<0.01),serum group containing herbs(0.4871±0.0195)compared with Ox-LDL group,there's difference(p<0.05),implied DuiGui BuXue Decoction could downregulate TNF-α's protein activity.3.ICAM-1's WB results:Ox-LDL group protein density area ratio is 1.0169±0.0128, compared with control group(0.2365±0.0113),the difference had great meaning (p<0.01),implied after Ox-LDL stimulate RAW264.7 cells,ICAM-1's protein activity obviously.Inhibitor group(0.5265±0.0232)compared with Ox-LDL group,difference was obvious(p<0.01),serum group containing herbs(0.6791±0.0164)compared with Ox-LDL group,there's difference(p<0.05),implied DuiGui BuXue Decoction could downregulate ICAM-1's protein activity.Conclusions:After Ox-LDL stimulated RAW264.7 cell,TNF-αand ICAM-1's protein activity obviously,DuiGui BuXue Decoction could downregulate TNF-αand ICAM-1's protein activity.As is pathologic process of the chronic inflammation,it develops to accompany with the inflammation always,and DuiGui BuXue Decoction maybe regulate and control relate inflammatory factors' expression to effect on As developing,so to decrease TNF-αand ICAM-1's protein expression may be one of mechanisms of DuiGui BuXue Decoction's action against As injury.
