Objective To establish an animal model of chronic obstructive pulmonary disease(COPD)to study the effects of traditional Chinese medicine Fufei Gushen Decoction on interleukin-17A(IL-17A),matrix metalloproteinase(MMP-9)and its specific inhibitor metalloproteinase inhibitor 1(TIMP-1).And to study its effects on airway inflammation and airway remodeling in COPD rats and its prevention and treatment mechanism.Methods A rat model of COPD was established by airway perfusion with smoke and lipopolysaccharide(LPS).50 Wistar rats were randomly divided into 5groups: normal control group,COPD model group,Fufei Gushen Decoction high,medium and low dose groups.The model was continued for 28 days.During the period,the mice were fed with drugs for 28 days.Theindicators were tested after 28 days.The general conditions of the rats ineach group were observed.The pathological changes of HE staining wereobserved.The expression of IL-17 A in serum and alveolar lavage fluid ofeach group was detected by ELISA after the collected arterial blood andalveolar lavage fluid of rats were anticoagulated and centrifuged.Thelung tissue of rats was taken,and the lung tissue was homogenized andcentrifuged.The expression of MMP-9 and TIMP-1 in lung tissue wasdetected by ELISA.A Statistical method was performed to analyze the results.ResultsThe results of HE staining showed that the cilia of the columnar epithelial cells fell or even fell off.The goblet cells increased and a large number of inflammatory cells secreted.The secretion of the lumen increased and the mucosal tissue became congested and edema.The alveolar wall became thin and even partially ruptured into a large pulmonary vesicle.The ELISA results show that the expressions of IL-17 A and MMP-9 in the model group were higher than those in the normal group(P<0.05),while the expression of TIMP-1 was significantly lower than that in the normal group(P<0.05).The expression of IL-17 A in serum and alveolar lavage fluid of rats in Fufei Gushen Decoction Groups was lower than that in model group(P<0.05).The expression of MMP-9 in lung tissue of rats in Fufei Gushen Decoction Groups was lower than that of model group(P<0.05).The expression of TIMP-1 in lung tissue of rats in Fufei Gushen Decoction Groups was higher than that of the model group(P<0.05).Among them,the middle dose group had the most obvious treatment effect,and the high dose and low dose groups were slightly weaker.Conclusion1.Airway inflammation of COPD may be related to the change of IL-17 A level.Airway remodeling of COPD may be related to the dysequilibrium of MMP-9/TIMP-1.2.The mechanism of Fufei Gushen Decoction in the treatment of COPD maybe is that it can reduce the levels of IL-17 A /MMP-9 while increase the level of TIMP-1,reduce airway inflammatory response and slow airway remodeling,thus it can achieve the effects of COPD prevention and treatment.
