Objetives:To investigate the effect of Fufei gushen decoction on the content of SIgA in the bronchoalveolar lavage fluid and the t-lymphocyte subgroup in the lung tissue of COPD rats,and to provide a new idea for the clinical prevention and treatment of COPD,and to provide a scientific basis for the Future development and utilization of fufei gushen decoction for the prevention and treatment of COPD.Methods: Fifty healthy and clean male Wistar rats were randomly selected as the blank control group after 1 week of adaptive feeding,and the remaining 40 rats were used as the model group.All rats in the model group were made into COPD model by means of fumigation combined with intratracheal injection of lipopolysaccharide for 28 days.On the 29 th day,2 model group rats and 2 blank control group rats were randomly sacrificed for HE staining,and the morphological changes of lung tissues were observed under light microscope and compared to evaluate the success of COPD model establishment.The rats were randomly divided into model group,high dose group,medium dose group and low dose group.Thereafter,the blank control group and model group were given distilled water by gavage,and each dose of traditional Chinese medicine group was given corresponding concentration of traditional Chinese medicine decoction by gavage twice a day for 28 days.Observe the appearance and living state of rats;The morphological changes of lung tissue and the effects of different doses of fufei gushen decoction on the pathological changes of COPD rats were observed under the microscope.SIgA in right bronchoalveolar lavage was detected by ELISA.Immunohistochemistry was used to detect the positive expression rate of T lymphocyte subsets in left lung tissue(mainly CD4+ and CD8+).Results:1.General information: the blank control group rats had shiny hair,quick reaction,normal activity,stable and even breathing,and rapid weight gain;COPD model group rats had withered hair,yellow hair,slow reaction,little movement and curl up,shortness of breath,cough and sneeze,increased oral and nasal secretions,slow weight gain,little food intake,and dry stools.Different doses of fufei gushen decoction of traditional Chinese medicine group above have different degrees of improvement.2.Under light microscope,HE staining results of lung tissue showed that there was no abnormality in bronchial mucosa,cilia and alveoli of rats in the blank control group.The bronchial epithelial structure of the rats in the model group was destroyed,with a large number of inflammatory cells infiltrating,cilia adhesion partial shedding,thin alveolarwall and large pulmonary vesicles forming.The morphological observation of lung tissue in different doses of fufei gushen decoction showed different degrees of improvement.3.The SIgA content in the right bronchoalveolar lavage fluid(BALF)of rats in each group showed that the SIgA content in the model group was lower than that in the blank control group,and the difference was statistically significant(P<0.01).The high,medium and low dose groups of fufei gushen decoction were all higher than the model group,and the difference was statistically significant(P<0.05).The medium dose group of fufei gushen decoction was higher than the high and low dose group,and the difference was statistically significant(P<0.05).4.Each group rats left T cell subsets in lung tissue and cell subsets(CD4 +,CD8 +)positive results show that: the model group was higher than the blank control group,the difference was statistically significant(P < 0.01).The high,medium and low dose groups of fufei gushen decoction were all lower than the model group,and the difference was statistically significant(P<0.05).The medium dose group of fufei gushen decoction was lower than the high dose group and the low dose group,and the difference was statistically significant(P<0.01).Conclusions:1.Fufei gushen decoction of traditional Chinese medicine can improve the general situation of COPD rats and pathological changes of lung tissue.2.Fufei gushen decoction of traditional Chinese medicine can increase the content of SIgA in BALF of COPD rats and enhance the local mucosal immunity of the respiratory tract,thus reducing the number of acute COPD attacks.3.Fu lung solid renal drink Chinese traditional medicine can reduce T cell subsets in lung tissue and cell subsets(CD4 +,CD8 +)the positive expression rate,thus reduce the lung tissue inflammation factors secreted by CD4 +,CD8 + level,reduce the lung tissue damage,and delay the progress of disease in COPD.
