Effect Of Fufei Gushen Yin On Glucocorticoid Receptor Level In COPD Rats

Objective:To investigate the effects of Fufei Gushen Yin on the levels of glucocorticoid receptor(GR)in the lung tissue of rats with chronic obstructive pulmonary diseases(COPD).The mechanism of action of drinking on COPD.To provide scientific basis for the prevention and treatment of COPD in the prevention and treatment of COPD in the future,and provide reference for Chinese medicine to prevent and treat COPD.Methods: Fifty healthy male Wistar rats,8 weeks old,weighing200±20 g,were fed for 1 week.They were randomly divided into 10 blank control groups and 40 model mice.The model was made by intravesical instillation of Lipopolysaccharide LPS.Two rats died during the modeling period.After the 28 th day of modeling,2 rats were randomly sacrificed in the model group and the blank control group.The whole lung tissue was taken to the pathology department for pathological examination to confirm the success of modeling.After confirming the successful modeling,the rats were randomly divided into model group,high-dose group of Fufei Gushen Yin,middle dose group of Fufei GushenYin and low dose group of Fufei Gushen Yin Group 9 only.The high,medium and low doses of Fufei Gushen Yin were administered with1.02 g Chinese medicinal formula granules/100g/day,0.51 g Chinese medicinal formula granules/100g/day,0.26 g Chinese medicinal formula granules/100g/day.The blank control group and the model group were administered with distilled water,1 ml/100 g/day,and administered twice a day,3 ml each time.After the 28 th day of the drug administration,all the rats were sacrificed,and the right upper lung lobe was quickly extracted and placed in a 10 ml centrifuge tube,frozen in liquid nitrogen,and placed in a-80 ° C refrigerator for treatment.The remaining right lobe was fixed in 4% tissue cell fixative for 12 h,embedded in paraffin,and sectioned for HE staining and immunohistochemistry.The pathological changes of lung tissue were observed by HE staining.The expression of GR in lung tissue was detected by immunohistochemistry.The expression of GR mRNA in lung tissue was detected by real-time fluorescent quantitative polymerase chain reaction(RT-PCR).Results:(1)Pathological results of HE staining of lung tissue sectionsThe bronchial wall structure of the lungs of the blank control group was almost intact,no obvious damage,only a few infiltration of inflammatory cells,no congestion and edema of the mucosa,complete structure of the alveolar space,no obvious damage and fusion.In the lungtissue of the model group,the epithelial cilia adhesion and even shedding were observed,and the bronchial wall was severely damaged.A large number of inflammatory cell infiltration was gradually infiltrated into the alveoli by the trachea,the tracheal wall was thickened,and the alveolar wall structure was damaged seriously or even thinned and broken.Alveolar fusion results in the formation of pulmonary bullae,consistent with the pathological features of COPD.After 28 days of treatment with Fufei Gushen Yin,the pathological changes of lung tissue in the high,middle and low dose groups of Fufei Gushen Yin were significantly reduced compared with the model group,such as bronchial wall thinning,inflammatory cell infiltration and congestion and edema.The situation was significantly alleviated,and the expansion of the alveolar cavity was reduced.The improvement of the middle dose group of Fufei Gushen Yin was the most obvious.(2)Results of immunohistochemical detection of GR expression in lung tissue of rats in each groupCompared with the normal group,the expression of GR in the lung tissue of the model group was significantly decreased((49)< 0.01).Compared with the model group,the expression of GR in the lung tissue of the high,middle and low dose groups of Fufei Gushen Yin was significantly increased((49)<0.01).Compared with the high dose and low dose of Fufei Gushen Yin,the expression of GR in the lung tissue ofFuzhong Gushen Decoction group was significantly increased((49)<0.05).There was no significant difference in the expression of GR between the high-dose and low-dose groups of Fufei Gushen Yin((49)>0.05).(3)RT-PCR detection of the expression of GR mRNA in lung tissue of rats in each group by Fufei Gushen YinCompared with the normal group,the expression level of GR mRNA in the lung tissue of the model group was significantly decreased(P<0.01).Compared with the model group,the expression of GR mRNA in the lung tissues of Fuzhong Gushen Decoction group was significantly increased((49)<0.01).Compared with the high dose and low dose of Fufei Gushen Yin,the expression of GR mRNA in the lung tissue of Fuzhong Gushen Decoction group increased significantly((49)<0.05).There was no significant difference in the expression of GR mRNA in lung tissue between high dose group and low dose group of Fufei Gushen Yin((49)>0.05).Conclusion:(1)Fufei Gushen Yin can up-regulate the GR level of lung tissue in COPD rats.(2)Fufei Gushen Yin can increase the effect of COPD by up-regulating the level of GR and increasing the binding rate of GC to GR.
