Objective: To investigate the effect of Qingnao Yiyuan Decoration on the expression of MBP and NF 200 in rats by acute chemise brain injury,and to explore the possible cerebral protection mechanism of Qingnao Yiyuan Decoration.Methods: 192 healthy male SD rats were randomly divided into 4 groups by random number table,the normal control group(12 animals),the sham operation group(60 animals),the chemise injury mode group(60 animals),and Qingnao Yiyuan group(60 animals).Except the normal control group,all the remaining groups were further divided into 5 subgroups according to the five time points of 1d,3d,7d,14 d,28d,after chemise injury,and each subgroups had12 rats in it.The following methods were used to made: the rats in the normal control group were feed and dinkie normally;the rats in sham operation group were operated with MCAO,but used 8mm fishing line inserted into blood vessel that could not enough to block middle cerebral artery blood flow,the rats were irrigated stomach by same volume of distilled water after being awake until the rats were euthanize;the chemise injury mode group was given the same volume of distilled water on seven continuous days before MCAO,after operation,same volume of distilled water was continued to be administered to the stomach until rats were killed at different stages;the Qingnao Yiyuan group was narcotized and operated the same methods with chemise injury mode group,also they were given the same volume of Qingnao Yiyuan defection seven continuous days before MCAO,when the operation was completed,the rats were irrigated stomach by Qingnao Yiyuan defection after being awake until the rats were killed at different stages.the Longa 5 like-rt scale system was used to score neurological defect symptoms in rats.Specimens of each group were collected at 1d,3d,7d,14 d,28d.Observed infarct volume of chemise brain tissue by TTC staining.Observation of pathological changes of neurons in chemise brain tissue by HE staining.Detection of MBP mRNA expression in chemise brain tissue by RT-PCR,and the expression of NF200 protein was measured by Western-blot.Results:(1)Neural function defect score: the scores of neurological deficits in all rats of normal control group and sham operation group were o,both the chemise injury mode group and Qingnao Yiyuan group had different degrees of neurological deficits,compared with the chemise injury mode group,the score of neurological impairment in the Qingnao Yiyuan group was lower(P<0.05).(2)The results of detection on cerebral infarction volume by TTC staining: gray-white infarction was not found in the normal control group and the sham operation group.both the chemise injury mode group and Qingnao Yiyuan group showed different sizes of gray-white chemise infarction.Compared with the chemise injury model group,the range of cerebral infarction in Qingnao Yiyuan group decreased.(3)The results of HE staining: the neurons in the normal control group and sham operation group were normal in morphology,clear and complete in structure,without pathological changes.After the chemise brain injury,the nerve cell had necrosis and apotheosis,there was marked eutrophic infiltrate within and around the chemise brain,nucleus deformity finally disappear in the cell,in the cytoplasm the binary actualization was extensive,and nest mass necrosis was visible.at the same time point,Compared with the chemise brain injury mode group,the Pathological changes in Qingnao Yiyuan group were significantly alleviated.Necrosis of nerve cells was reduced,the arrangement of nerve cells was irregular,and edema in chemise area was obvious.also eutrophic infiltration were seen around the nerve cells,there were no micro vacuole formation and nest necrosis found in the cytoplasm.(4)The results of RT-PCR showed that there was no significant difference in MBP mRNA expression between the normal control group and the sham operation group at the same time point(P>0.05);Compared with sham operation group,the expression of MBP mRNA in chemise injury model group decreased significantly at different time points(P<0.01);Compared with the chemise mode group,the expression of MBP mRNA in the Qingnao Yiyuan group was significantly increased at each time point(P<0.05).(5)The results of Western-blot showed that there was no significant difference in the expression of NF200 protein between the normal control group and sham operation group at the same time point(P>0.05).After operation,the expression of NF200 in cerebral alchemist injury mode group was decreased significantly(P<0.01).compared with the chemise injury model group,the expression of NF200 in the Qingnao Yiyuan group was significantly increased at all time points.Conclusion: The play a euro-protective role in rats with cerebral alchemist injury,its possible mechanism was increased the expression of MBP and NF200.
