Objectives: To investigate the effect of Qingnao Yiyuan Decoction on the cerebral cortex and expression of Hippocampal c-jun and ATF2 after focal cerebral ischemia/rep-erfusion(I/R)in rats and to discuss its mechanism of nerve protection.Methods: One hundred and ninty-two healthy SD rats were randomly divided into two big groups by random number table.Ninty-six rats were randomly divided into four subgroups in each group: the normal control group(group A6 animals),sham operation group(group B 30 animals),model group(group C 30 animals)and Qingnao Yiyuan group(group D 30 animals).The groups of B?C?D were further divided into five subgroups(n=6)according to the ischemic injury after 1d,3d,7d,14 d and 28 d.The following processing was made: the rats in group A were feed and drinking normally,also given same volume of distilled water;the rats in group B were narcotized and operated with MACO but ues 8mm Surgical line.after same volume of distilled water was given seven days,,also feed and drinking normally after operation and given same volume of distilled water;the rats in group C were narcotized and operated with MACO after same volume of distilled water was given seven days,the rat model of acute cerebral ischemia injury in rats were made after operation,the rats were irrigated stomach by physiological saline after being awake until the rats were euthanized;the rats in group D were operated the same methods with group C after irrigated stomach by same volume of Qingnao Yiyuan seven days,the rats were irrigated stomach by Qingnao Yiyuan after being awake until the rats were euthanized.Scores of degree of defect in neurological function in rats were made according to the Longa 5 like-rt scale.The specimen in group B,C and D were collected on different phase accordingly.The expression of c-jun in the hippocampal on the ischemic side cortex of rats were measured by immunohistochemical SP method in one group,and the expression of ATF2 on the ischemic side of in hippocampus were measured by Western Blot in the other group.Results: 1.Results of Neurological deficit score: The scores of neurological deficits in all the rats in the blank control group and sham group were 0,and both the model control group and the Qingnao Yiyuan Decoction group had different degrees of neurological deficits.Compared with the model control group,the score of neurological impairment in the Qingnao Yiyuan Decoction group was lower,and the differencewasstatistically significant(p<0.05).2.Results of Immunohistochemical SP method test: Compared with the sham operation group,the expression of c-jun in the brain tissue of the model group and the Qingnao Yiyuan Decoction group was significantly increased at all time points,the difference was statistically significant(p <0.05).Early(first day)showed a clear upward trend,then(the third day)began to decline.Compared with the model group,the positive rate of c-jun in Qingnai Yitong group was significantly lower at all time points,the difference was statistically significant(P <0.05).Early(first day)showed a clear upward trend,then(the third day)began to decline.3.Results of Western Blot: Compared with the sham operation group,the expression of ATF2 in model group and Qingnao Yiyuan Decoction group increased significantly at each time point,the difference was statistically significant(p <0.05).Compared with model group,the expression of ATF2 in Qingnao Yiyuan Decoction group was significantly lower at all time points,the difference was statistically significant(P <0.05).Conclusion: Qingnao Yiyuan Decoction can reduce the expression of c-jun and ATF2 after cerebral ischemic injury in rats,which ma y be one of the mechanisms of its effect on neuronal protection af ter cerebral ischemia.
