Objective: To explore the dominant differences in the community structural diversity of oral salivary bacteria among children aged 3~5years old in Mongolian,Uygur and Han children in Bozhou,Xinjiang.Methods : The 120 cases of aged 3 ~ 5 years children saliva samples(40 Han,40 Uygur,40 Mongolian)were randomly selected from the data sample database established by the oral epidemiological survey in Bozhou,Xinjiang.In each ethnic groups,there were 20 severe early caries(ECC)samples and 20 caries-free(CF)samples,taking non-irritating saliva,and extracting bacterial DNA for polymerase chain reaction(PCR)amplification and denaturing gradient gel electrophoresis.(DGGE)analysis,finally using the Quantity One software to analyze the PCR-DGGE fingerprint.Results: Analyzing the DGGE profile,we found that salivary bacteria had ethnic differences and intra-individual differences;the total band numbers and diversity index of Uygur population(band numbers: 87.00±10.40,diversity index: 5.99±1.12)was greater than Han nationality(band numbers: 63±8.07,diversity index: 3.29±0.74)and Mongolian(band numbers: 71.6±5.79;diversity index: 4.98±0.50)(P<0.05),the bacterial community structure diversity of severe early caries group was less than the caries free group(P<0.05);There were ethnic differences in the bacterial structure of the caries free children group(P<0.05).The cluster analysis found most of the samples in the ethnic groups showed obvious clusters.Conclusion: Bacterial community structure of oral saliva of young children has ethnic differences in Bozhou area.During early caries development,bacterial diversity in the saliva likely decreases.
