OBJECTIVE:Dental caries so far is still human the most common chronic infectious diseases, because of its high incidence and wide-ranging, the harm is self-evident. Children are caries incidence populations, and difficult to treat, and easy to affect the growth and development of children, the harm is more prominent.Caries is typical of infectious diseases caused by bacteria, but more and more evidence that the incidence of dental caries is the ecological balance of oral microbial populations and cause tooth surface is broken, not a single one or several dental caries bacteria at work, for this study the process of change microbial populations caries, clear process to occur caries and caries etiology has a very important significance.The second-generation sequencing technology for in-depth understanding of the structure and diversity of microbial populations have very obvious advantages, the present study is the use of Illumina MiSeq sequencing technology, comparative analysis of the structure and diversity of different degrees of caries in children and non-caries plaque microbial populations, expect children caries associated microflora, and microbial populations in four different caries susceptibility of children on dental plaque bacteria enrichment analysis, so as to understand changes in microbial populations in caries, so providing a biological caries preventive measures for the prevention and treatment.METHODS:2015 years, Dezhou City nursery randomly selected 135 children under 6 years old, and its inspection caries plaque samples were collected, divided into no caries, caries low, moderate caries, caries height, four group. Four groups each randomly selected sample of 10 6-year-old dental plaque of children included in the study, plaque samples obtained after routine sampling extracted DNA, utilizing Illumina MiSeq sequencing method, gene sequencing for 16SrRNA V4 variable region designed universal primers in each group structure and diversity of oral microflora.RESULTS:Using Illumina MiSeq sequencing methods to obtain a total of 516,248 pieces sequences effective sequence pretreated statistical quality control and get to 478,272, an average of 43021. It identified 21 species and 238 kinds of bacteria belonging to the genus door. Microbiota between the groups have a greater similarity, there are five main microbial flora dominant bacteria Doors:Firmicutes, Proteobacteria, fusobacteria, Bacteroides, and Actinobacteria. The analysis found that there are seven major dominant species:Leptotrichia, Streptococcus, Prevotella, Corynebacterium, Veillonella, Clostridium genus, Fusobacterium, Parascardovia, in the occurrence of dental caries in a dominant position, thereby displaying the development of these species and caries should show some correlation.CONCLUSION:1 different levels of dental caries of children dental plaque microbial populations have relatively large similarity, which proves the correctness of the ecological plaque hypothesis.2 increased with the severity of caries, does have some microbial populations in the occurrence of certain changes, this change and caries occurrence of concomitance, which also need to further study3 we study for dental caries of in-depth understanding of the microbial etiology, further validation of the ecological plaque hypothesis, laid the foundation.
