Objective:Based on the clinical experience of the tutor in the treatment of post-stroke depression,the important therapeutic principles of "shu gan jie yu,jian pi yang xue" were expounded systematically.Meanwhile,the effect of paeoniflorin,an effective component of paeoniae,on poststroke depressed rats and the possible mechanism of action were explored through the experimental study on the establishment of drug use experience by the tutor.Methods:The MCAO rat models were duplicated by the MCAO and randomly divided into four groups:Sham,PSD,Paeoniflorin,and Fluoxetine.Two days after the operation,the PSD,Paeoniflorin and Fluoxetine groups were given CUMS combined with isolated feeding for three weeks to copy the PSD models.The intraperitoneal injection was performed the first day of CUMS.The Neurological Function Score and Beam Walking Test Score were used to evaluate the degree of neurological deficit.The state of depression was evaluated by the Open Field Test and Sucrose Preference Test.The expression level of BDNF and p CREB protein in hippocampal neurons was detected by the methods of Western-Blot and Immunofluorescence.Results:1.Neurological Function Score:The Sham group scored lower than the others one day after the MCAO(P<0.05),while the scores of the Paeoniflorin group were significantly lower than the PSD group three weeks after the CUMS(P<0.05),and were close to the Sham and Paeoniflorin groups(P>0.05).2.Beam Walking Test Score:The Sham group scored lower than the others one day after the MCAO(P<0.05).Three weeks after the CUMS,the scores of the PSD group were higher than the others(P<0.05),the scores of Paeoniflorin and Fluoxetine groups were higher than the Sham group(P<0.05),but there was no significant difference between them(P>0.05).3.Sucrose Preference Test:The sucrose consumption rate was similar in all groups after the MCAO and one week after the CUMS(P>0.05).The consumption rate of the Paeoniflorin group was significantly higher than the PSD group two weeks after the CUMS,and was similar to the Sham and Fluoxetine groups(P>0.05).4.Open Field Test Score:There was no significant differences between the horizontal and vertical movement scores of all groups before surgery(P>0.05).Three weeks after the CUMS,the horizontal and vertical movement scores of the PSD group were significantly lower than the others,and there was no significant differences in the vertical movement scores among the Sham,Paeoniflorin and Fluoxetine groups(P>0.05).The horizontal movement scores of Sham group were higher than that of Paeoniflorin and Fluoxetine groups(P<0.05)but there was no significant differences between the last two(P>0.05).5.Western Blot:The expression of BDNF and p CREB protein in the PSD group was lower than the others(P<0.05),and there was no significant difference among the Sham,Paeoniflorin,and Fluoxetine groups(P>0.05).6.Immunofluorescence The positive expression of BDNF and p CREB protein was increased and the Fluorescence intensity was significantly enhanced in the Paeoniflorin group compared with the PSD group,and was similar to the Sham,Paeoniflorin,and Fluoxetine groups.Results:1.Paeoniflorin can promote the neurofunctional recovery of PSD rats and improve the depressive state;2.Paeoniflorin can improve the expression of BDNF ? p CREB protein in the hippocampal neurons of PSD rats;3.The therapeutic effect of Paeoniflorin on PSD rats was similar to the Fluoxetine.
