In this study,we first conducted literature surveys and field surveys of Dioscorea opposita(produced in Jiaozuo,Henan Province).We found that the main varieties planted and circulated were Dioscorea opposita Thunb.cv.Taigu and Dioscorea opposita Thunb.Cv.Tiegun),this article is based on Taigu Yam and Tiegun yam as the research object,and evaluates the quality of Chinese yam medicinal materials processed by different processing methods,so as to select the optimal production process of yam;and this research collects Huai yam in different periods.Chemical analysis was conducted to find out the law of accumulation of chemical components in the growth process of Dioscorea opposita and provide theoretical basis for rational harvesting.The main research content is divided into the following three chapters:The first chapter is the investigation of the research status of the processing technology of the drug production area in Huaishan.Through consulting ancient herbal medicine,we learned that yam processing is mostly used for drying and steaming in ancient times.In modern times,with the advancement of economy and science and technology,the processing of yam has become more diversified.The traditional processing technology of producing areas is sulfur smoky processing,which is processed into "Guang yam" and "Mao yam".Modern non-sulfur processing methods include Electric blast drying,far-infrared drying,vacuum drying,freeze-drying and microwave drying,etc.,processing products are mostly "hill tablets." It is known from the survey of origin that the current sulphur-smoke processing by farmers is based on experience.There is no scientific normative sulphur-smearing process as a guideline,and there is a problem that the sulphur dioxide residue is excessive.Therefore,it is necessary to develop a standardized and reasonable production process of yam.The second chapter is the research on the processing technology of yam medicine.On the basis of literature review and origin survey,the samples collected from this study were mainly from farmer processing,enterprise processing,and laboratory simulation.The locations of farmer processing include Wenxian,Wuzhi,Anguo,and Pingyao.The samples processed by the company were from two enterprises in Wenxian and Wuzhi in Henan Province,and the laboratory simulated processing included sulphur processing samples and non-sulfur processing samples.The quality of samples collected and processed was evaluated,and water-soluble extracts,polysaccharides,allantoin,adenosine,monosaccharides,and oligosaccharides were used as chemical evaluation indicators,and moisture,ash,and heavy metals were detected at the same time.The results of variance analysis and principal component analysis were finally performed using SPSS 20.0.The results showed that in the laboratory simulated non-sulfur processing process,the electric blast drying process was the best,the temperature was 80?;the laboratory simulated sulfur fume process,Sealed and smoked,leaving 2cm wide vents on both sides,the amount of 2g of sulfur per kilogram of yam smoked the most suitable;collected Henan Wenxian,Wuzhi,Anguo,Pingyao and other places of light yam,Mao yam,Shanxi Pingyao smoky yam due to the short waterfront time,the overall content of its chemical composition than other regions,but the processing process is not mature,Henan Wenxian,Wuzhu processing more mature;between non-sulfur dried fruits and fresh products Comparing the results,the processing technology of the enterprise is more stable and the quality is better than that of the farmers.The third chapter is the study on the law of accumulation of chemical components of Dioscorea opposita.This study began on September 15 when yam growth began,and was collected every 15 days.Taigu yam was sampled in parallel for 4 batches.Tiegun yam was sampled in parallel for 3 batches.The yam growth and maturation was stopped by sampling by farmers.The collected yam was examined for moisture,ash,and heavy metals in accordance with the 15th edition of Chinese Pharmacopoeia,and the contents of water-soluble extracts were determined.The phenol-concentrated sulfuric acid method was used to determine the yam polysaccharide content,and high performance liquid chromatography was used for yam medicine.The contents of allantoin,adenosine,monosaccharides,and oligosaccharides were determined.According to the general principles for determining the suitable harvesting period of medicinal materials,the combined analysis of the dynamic accumulation rules of yam yield and chemical composition shows that:Taigu yam is suitable for excavation in mid-December,combined with production factors,it can also be excavated in winter.However,it is recommended that excavation be completed at the end of March of the following year to avoid reduction in the yield and content of active ingredients;Tiegun yam in the middle of December that year better excavation.
