Achyranthes bidentata was used as the research object In this paper.Through the determination of the contents of Achyranthes bidentata in different harvesting periods,The change law of dynamic accumulation of Achyranthes bidentata in different harvest periods was investigated to obtain the best harvest period of Achyranthes bidentata roots.At the same time,through the study of laboratory simulated processing samples,processed samples of processing areas of farm households,and processed samples of enterprises,the differences between different processing technologies were compared to optimize the best processing technology for Achyranthes bidentata.Finally,establish fingerprints of Achyranthes bidentata,Comprehensively evaluate the intrinsic quality of Achyranthes.The first part is the chemical composition accumulation change of the Achyranthes bidentata at different harvest time.This part of the experiment studied the samples from three plots at seven different harvest periods.The moisture,ash and heavy metals of Achyranthes bidentata were tested.The experimental results showed that the contents of moisture,ash and heavy metals were within the limits.At the same time,the content of chemical constituents such as the dryness rate,alcohol-soluble extracts,polysaccharides,oleanolic acid,?-ecdysterone,25R-inokosterone,25S-inokosterone is taken as the Achyranthes bidentata different harvest sample evaluation index.The topsis evaluation method in DPS data processing system was used to comprehensively evaluate the chemical quality of Achyranthes bidentata in different harvest periods,and the best harvest period of Achyranthes in the period from mid-November to early December was concluded.This conclusion is consistent with the traditional harvesting of producing areas.The phases are consistent,indica-ting that the harvest time of the Achyranthes in the origin is scientific and reasona-ble.In addition,Used ICP-MS determined the content of 22 inorganic elements in different periods of Achyranthes bidentata,to compared the difference of inorganic elements content in different periods of Achyranthes bidentata.Used SPSS 20.0 statistical software to analyze the variance of inorganic elements(excluding heavy metal elements Pb,Cd,As,Hg,Cu)in Achyranthes bidentata in different harvest periods.The results showed that the contents of Cr,Ni,Zn,Se,Ba,Al,and P in the Achyranthes bidentata had no significant difference in seven different harvest periods,and Li,Na,Mg,K,Ca,Ti,Mn,Fe,Co,and Sr elements were significant difference during each harvest period.The second part is the study of processing technology of Achyranthes bidentata,including processing at the laboratory,processing at the place of origin and process-ing at the enterprise.All processing techniques were evaluated using alcohol-soluble extracts,polysaccharides,oleanolic acid,?-ecdysterone,25R-inokosterone,25S-inokosterone as indicators to evaluate the differences between the processing technologies.Laboratory processing is divided into four aspects,The first section is use orthogonal test to investigate the effect of different oven,different drying temperature and the different state of drying on the quality of Achyranthes bidentata.The results of the determination of each component were analyzed using the AHP combined with the comprehensive scoring method,The optimum integrated proce-ssing technology of the Achyranthes bidentata is to cut the long section of the fresh Achyranthes bidentata 1.0-1.5cm and dry at 60 degrees centigrade.The second section is selecting the processing technique for cutting the Achyranthes bidentata in different water content.The measurement results of sample are evaluated using the principal component comprehensive score method.The results showed that Achyranthes bidentata cut at the moisture content of 20%-30%,this processing technology ranked first,and the appearance of the sample under the process of the best performance.The third section is to examine the complete branches,long section and short section of fresh Achyranthes bidentata under different power in vacuum microwave drying.Using the analytic hierarchy process combined with the comprehensive scoring method to evaluate the results of each component,the results show that the sample processed at 3KW power is better.The fourth section is the effect of four different processing methods on the quality of Achyranthes bidentata by comparing the electrothermal dryness,far infrared drying,vacuum drying,vacuum microwave drying and so on.Finally,the conclusion is that the best processing method using electrothermal blast drying is concluded.The results showed that there was no significant difference between dry products and fresh products by comparing the chemical components of Achyranthes bidentata collected from different areas of Henan Wuzhi,Henan Wenxian,Hebei Anguo and Inner Mongolia Chifeng.ANOVA was used to compare the Achyranthes bidentata complete branches and decoction pieces collected by different companies.The results showed that there was no significant difference between complete branches and decoction piece in the chemical composition of Achyranthes bidentata.The contents of moisture,ash and heavy metals in different processed Achyranthes bidentata samples were detected,The results showed that moisture,ash and heavy metal contents were all within the limit range.ICP-MS was used to determine 22 inorganic elements in Achyranthes bidentata from different areas,and the results of inorganic elements(except for heavy metal elements Pb,Cd,As,Hg,Cu)were analyzed by ANOVA.The results show that there is a significant difference in the content of inorganic elements between different producing areas.At the same time,using cluster analysis to analyze the inorganic elements from different producing areas,the samples of Henan Wuyi and Wenxian of Henan gathered into one category,and the samples of Hebei Anguo and Chifeng of Inner Mongolia clustered into one category.It can be seen that the geographical distribution is significant.The third part is to establish a fingerprint of Achyranthes bidentata.A total of 14 common peaks have been calibrated,and three known compounds were identified.The similarity between the sample Fingerprints and the reference fingerprintis above 90%.It shows that the generated reference fingerprintis has high reliability,The quality of the Achyranthes bidentata can be described and evaluated by the reference control fingerprint.
