Objective To investigate the apoptosis eftect of the Black ginseng,Black garlic and their mixture on Hepatocellular carcinoma HepG-2 cells and its mechanism.Methods MTT assay was used to determine the effect of Black ginseng,Black garlic and their mixture on the proliferation of HepG-2 cells.The influence of cell apoptosis and cell cycle employed was tested by flow cytometry assay.Western Blot was used to detect the expression of Caspase-3,Bax,Bcl-2 in HepG-2 cells.Results MTT assay showed that the growth of HepG-2 cells was inhibited when the concentration of Black ginseng was 7.5?15g/L,the Black garlic is 10?15g/L,the mixture is 30 g/L(Black ginseng:Black garlic=1:2).The cells became swollen and round with unclear boundaries inverted by Microscopic Observation.The early apoptosis rate of HepG-2 cells was significantly increase with the extracts of Black ginseng,Black garlic and their mixture.The Black ginseng acted on inhibition was associated with arresting cell cycle at G1 and S phase and the Black garlic arresting cell cycle at G1 and G2 phase.Compared with the control group,the expression of Caspase-3 and Bax protein significantly increased,Bcl-2 protein significantly decreased treated by Black ginseng,Black garlic and their mixture.Conclusion Compared with the control group,the inhibition and apoptotic induction effects on HepG-2 cells Black ginseng,Black garlic and their mixture;the early apoptotic rate increased and cell arrest in G1 phase induced.Inonotus obliquus can up-regulate the expression of Bax/Bcl-2,Caspase-3 protein and down-regulate the expression of Bcl-2 protein.
