Purpose:To find out the molecular mechanisms of Aged Black Garlic Extract(ABGE)through regulating the TGF-?1/Smad4 signaling pathway on growth and metastasis of Panc-1 cells,and to further defined the signal transduction pathways and its mechanism of action of Aged Black Garlic Extract(ABGE)inhibit pancreatic cancer of growth and metastasis.Methods:In this study,we choose the Panc-1 cells as the object,and the research were divided into two parts,that is the vitro and vivo,respectively.In the vitro part:To detect the inhibition growth of rate of Panc-1 cells by CCK-8 methods;To detect the effect of ABGE on expression of TGF-?1 and MMP-9mRNA in melanoma Panc-1 cells by Realtime-PCR;To observe the expression levels of TGF-?1 and Smad4 of ABGE on Panc-1 cells with Western blot;To observe the invasion ability of ABGE on Panc-1 cells by Transwell methods;In the vivi part:To establish the Panc-1 mouse transplanted tumor model and the Panc-1 metastatic model.To observe the the inhibition effect of ABGE for growth of transplanted tumors and metastatic incidence on liver;To detect the expression of ABGE on TGF-?1 and Smad4 of transplanted tumors-with immunohistochemical methods.Results:(1)ABGE could reduce the activity of the Panc-1 cells,and inhibit the proliferation of Panc-1 cells significantly in vitro.(2)ABGE could up-regulate the expression of Smad4 and down-regulate TGF-?1 expression in Panc-1 cells in vivo,to modulation of TGF-?1/Smad4 signaling pathway,and could decrease the expression of TGF-?1 and MMP-9,in order to reduce movement ability of the cells,and inhibit the metastasis of cancer.(3)ABGE could inhibit the metastasis rate of the liver in vivo,and affect the related protein expression of tumor metastasis of the TGF-?1/Smad4 pathway.Conclusion:ABGE could up-regulate the expression of Smad4 and down-regulate TGF-?1 expression in Panc-1 cells in vivo,to modulation of TGF-?1/Smad4 signaling pathway,above may be the important role of the mechanism of the angiogenesis and metastasis of Panc-1 cells.
