Exploration Of Danggui Sini Decoction On Glucoseuria Based On NF-kappa B Signaling Pathway Protective Mechanism Of Peripheral Neuropathy Indiabetic Rats

Objective: By observing changes of NF-kappa B signaling pathway under intervention of Chinese medicine,reveals the correlation between the protective mechanism of Danggui Sini Decoction on diabetic peripheral neuropathy and the NF-kappa B signaling pathway.Methods: Male rats of SD were selected to establish diabetic peripheral neuropathy model by chemical inducer streptozotocin The rats were randomly divided into model group,high dose group,low dose group and metformin group.Twelve healthy rats in the same batch were set as normal group.After 4 weeks,rats were given orally and their body weight and blood sugar were measured regularly every week.The rats were sacrificed 12 weeks after intragastric administration.The effects of sciatic nerve conduction velocity,pathological changes of sciatic nerve,nuclear factor-kappa B in sciatic nerve,interleukin-6(IL-6)and tumor necrosis factor-alpha in serum were detected.Results: 1.Compared with the model group,the blood sugar of rats in each treatment group decreased significantly(P<0.05).2.Compared with the model group,the motor nerve conduction velocity and sensory nerve conduction velocity increased significantly in the treatment group(P < 0.05).Compared with the blank group,the morphology of sciatic nerve of rats in the other groups changed significantly.After the intervention of traditional Chinese medicine,the treatment groups increased significantly(P < 0.05).The morphology of sciatic nerve in group A was improved(P < 0.05).4.Compared with model group,the expression of NF-kappa B protein and RNA in sciatic nerve in each treatment group was significantly decreased,and the expression of Interleukin-6 and Tumor necrosis factor-alpha in serum was significantly decreased(P < 0.01).Conclusion: Danggui Sini Decoction can effectively reduce the blood sugar of DPN rats,inhibit the expression of NF-kappaB and down-regulate the expression of inflammatory factors Interleukin-6 and Tumor necrosis factor-alpha.It has a certain therapeutic effect on DPN,and its mechanism may be related to the anti-inflammatory effect of inhibiting the signal pathway of NF-kappa B.
