Objective:To investigate the effete of Zhenwu decoction on nuelear factor kappa B signaling pathway of unilateral ureteralobstruction rats, and to illustrate the mechanism of the Zhenwu decoction on delaying renal interstitial fibrosis.Method:Copy the model of renal interstitial fibrosis in rats with unilateral ureteral obstruction. The150male wistar rats were randomly divided into six groups:sham operation group(21), UUO group (21), Zhenwu decoction of low-dosage group (27), Zhenwu decoction of medium-dosage group (27), Zhenwu decoction of high-dosage group (27), Niaoduqing granules group (27).The rats were given daily gavage the first day after surgery.A part of rats in each group were killed respectively at2ã€3ã€4weeks after surgery.Put to death the day after the collection of24h urine detection of24urinary protein excretion, executed the same day to collect blood for serum creatinine (Scr), blood urea nitrogen (BUN) index detection.Take the obstructed kidney tissue specimens, HE staining under the light microscope to observe the pathological changes; the pathological changes and expressions of NF-κB, TNF-α MCP-1in renal interstitium were observed.Results:(1)From the appearance point of view, treatment group of Zhenwu decoction in the corresponding time points of the obstructed kidney enlargement and stagnant water level were lighter than UUO group.(2)Rats in each group at different time points24hours urinary protein excretion were no significant difference (P>0.05)(3)Scr and BUN levels of Zhenwu decoction groups and Niaoduqing granules group were lower than UUO group at each indicated time points(p<0.05. P<0.01); at the same time point, Scr, BUN level of Zhenwu decoction of medium-dose group wa slowest; the difference of Scr, BUN level between Niaoduqing granules group and Zhenwu decoction of medium-dose group was not statistically significant.(4)Immunohistochemical results and ELISA result showed:the expressions of NF-κB gradually increased with the extension of the time of obstruction, in UUO groupã€Zhenwu decoction groups and Niaoduqing granules group; the expressions of NF-κB in Zhenwu decoction groups and Niaoduqing granules group were significantly lower than UUO group at the corresponding time point(p<0.05, P<0.01); at the same time point, the expressions of NF-κB in Zhenwu decoction of medium-dose group was lowest; the difference between the expressions of NF-κB in Niaoduqing granules group and Zhenwu decoction of medium-dose group was not statistically significant at2w and3w, but at4w the difference was statistically significant.(5)Immunohistochemical results showed:the expressions of TNF-α gradually increased with the extension of the time of obstruction, in UUO groupã€Zhenwu decoction groups and Niaoduqing granules group; the expressions of TNF-α in Zhenwu decoction groups and Niaoduqing granules group were significantly lower than UUO group at the corresponding time point(p<0.05, P<0.01); at the same time point, the expressions of TNF-α in Zhenwu decoction of medium-dose group was lowest; the difference between the expressions of TNF-α in Niaoduqing granules group and Zhenwu decoction of medium-dose group was not statistically significant.(6) ELISA result showed:the expressions of MCP-1gradually increased with the extension of the time of obstruction, in UUO groupã€Zhenwu decoction groups and Niaoduqing granules group; the expressions of MCP-1in Zhenwu decoction groups and Niaoduqing granules group were significantly higher than UUO group at the corresponding time point(p<0.05, P<0.01); at the same time point, the expressions of MCP-1in Zhenwu decoction of medium-dose group was lowest; the difference between the expressions of MCP-1in Niaoduqing granules group and Zhenwu decoction of medium-dose group was not statistically significant.Conclusion:(1) The Zhenwu decoction could improve renal function of UUO rats through reducing serum creatinine, blood urea nitrogen level.(2) The Zhenwu decoction could reduce the pathological changes of kidney in UUO rats.(3) The Zhenwu decoction could delay the Progression of renal interstitial fibrosis, through decreasing the expressions of NF-κB〠TNF-α〠MCP-1of UUO rats. |
