ObjectiveTo observe the effect of Shenzhuzhiyangfang TaZi on chronic perianal eczema by EASI scoring method in rats with chronic perianal eczema;To detect the content of nuclear factor erythroid related factor 2(Nrf-2)protein,superoxide dismutase(SOD)and malondialdehyde(MDA)in rat skin lesions,and to explore whether Shenshuzhiyaofang can inhibit free radicals and improve chronic perianal eczema by regulating Nrf-2 signaling pathway;To detect the contents of TNF-alpha(TNF-α)and IL-1beta(IL-1β),and to explore whether Shenzhuzhiyangfang can regulate inflammatory factors and inhibit the inflammatory response of chronic perianal eczema.MethodsForty healthy SD male rats were randomly divided into blank control group,model group,wet stain group and Western medicine control group with 10 rats in each group.Except for the blank control group,the other groups used2,4-dinitrochlorobenzene(DNCB)solution to establish the rat model of chronic perianal eczema.After successful modeling,the blank control group and the model group were given saline soaking,the soaking group and the western medicine control group were given antipruritic prescription soaking and Compound dexamethasone acetate cream topical application,twice daily intervention.The changes of perianal skin of rats in each group were observedand the EASI score of ZhaoBian was used for reference.Detection of TNF-αand IL-1 β in Rat skin tissue by Enzyme-Linked ImmunoSorbent Assay(ELISA)method and Immunohistochemistry(IHC)method,Nrf-2 protein by Western Blot(WB),SOD by hydroxylamine and MDA by thiobarbituric acid(TBA).The mechanism of Shenzhuzhiyangfang TaZi in treating chronic perianal eczema was discussed.Results1.Compared with the blank control group,EASI score of model group,wet-stained group and the western medicine control group,expression of Nrf-2protein,content of TNF-α,IL-1β,MDA increased,SOD content decreased(P<0.05).2.Compared with the model group,EASI score in the model area of rats in the wet-stained group and the western medicine control group decreased(P<0.05),the content of TNF-α,IL-1β,MDA decreased,the expression of Nrf-2protein and the content of SOD increased(P<0.05).3.Compared with the western medicine control group,EASI score in the model area of rats in the wet-stained group had no significant difference(P>0.05).There was no significant difference in the expression of IL-1β,Nrf-2,the content of SOD and MDA in skin tissue(P>0.05),the content of TNF-αdecreased(P<0.05).Conclusions1.Shenzhuzhiyangfang TaZi can improve the skin lesions of erythema,papules,scaly and mossy-like in rats with chronic perianal eczema;2.Shenzhuzhiyangfang TaZi can up-regulate the expression of Nrf-2 protein,increase SOD content and reduce MDA content,suggesting that the therapeutic mechanism of Shenzhuzhiyangfang TaZi on chronic perianal eczema may be related to regulating Nrf-2 signaling pathway,inhibiting free radicals and improving the body’s antioxidant capacity;3.The decrease of TNF-α and IL-1β in the tissues of rats with chronic perianal eczema suggests that Shenzhuzhiyangfang TaZi might be able to reduce inflammatory factors and alleviate inflammation.
