Objective:To observe the effect of haima on the hyperplasia of mammary glands models in rats and mice.To investigate the effect on function characteristics and molecular mechanism of Bushenyang traditional Chinese medicine on the hyperplasia of mammary glands.To reveal the pharmacological and pharmacodynamic mechanisms action of haima in the treatment of hyperplasia of mammary glands.Method:Empirical method 1:The rat of hyperplasia of mammary glands model was established by ovarian preservation and intramuscular injection of estradiol benzoate and progesterone.Continuous administration for 35 days,the nipple height and diameter of each group of rats were measured before and the end of the experiment.The activity of rat was observed after the last gavage.The changes of nipple diameter and height in each group were recorded 2 hours later,blood was taken from the eyeball of the rat and serum was isolated.The levels of E2,P,FSH,LH,GnRH,PRL,T in serum by enzyme-linked immunosorbent assay?Elisa?;removed thymus,spleen,uterus,weighed wet weight and calculated organ index;then taked the breast tissue,thymus,uterus,ovary,spleen and fix it in 4%formaldehyde solution.The pathologicalchangesofbreasttissue,thymusandspleenwere observed.immunohistochemical method was used to measure the positive expression of ER and PR in breast tissueEmpirical method 2:The mice of of mammary gland,thymus and spleen.Results:Compared with the blank control group,the breast diameter and the nipple height of the model group were significantly increased.The wet weight index of thymus and spleen decreased significantly,and the uterine index increased significantly,which indicated that the thymus gland and spleen were atrophic and the uterus was enlarged.The serum levels of E2,FSH,GnRH,PRL,and T in the model group was significantly increased,the contents of P was significantly decreased,and the expression of ER and PR was significantly increased in the model group.Estradiol benzoate combined with progesterone induces sex hormone disturbance in the rat model of hyperplasia of mammary glands,which is consistent with the hormone symptoms of HMG model.Compared with the model group,the diameter of the breast and the height of the nipple in the positive group and the large,medium,and small dose groups of the haima significantly decreased.The diameter of the breast and the height of the nipples in the positive group,the large,medium,and small dose groups of the haima were significantly reduced.The index of thymus and spleen was significantly increased and the index of uterus was decreased in the positive group and he large,medium,and small dose groups of the haima.The levels of serum E2,FSH,GnRH,PRL,and T were significantly increased,and the levels of P were significantly decreased,the LH content was increased significantly in the large and middle dose groups,while in the small dose group,the LH content was significantly increased in the small dose group.and the pathological changes of breast tissue,thymus,and spleen were improved.Compared with the blank control group,the breast diameter and the nipple height of the model group were significantly increased.The wet weight index of thymus and spleen decreased significantly,and the uterine index increased significantly,which indicated that the thymus gland and spleen were atrophic and the uterus was enlarged.This phenomenon is consistent with the appearance of thymus,spleen and uterus in hyperplasia of mammary glands animal model.The serum levels of E2,FSH,GnRH,PRL,and T in the model group was significantly increased,the contents ofLH and P was significantly decreased,Estradiol benzoate induces sex hormone disturbance in the mice hyperplasia of mammary glands model and is consistent with hormonal symptoms in the hyperplasia of mammary glands model.Compared with the model group,the diameter of the breast and the height of the nipple in the positive group and the large,medium,and small dose groups of the haima were significantly reduced;the thymus and spleen index of the mice increased,and the visceral index of the uterus was decreased,Serum E2,FSH,GnRH,PRL,and T levels significantly reduced and the levels of P significantly improved,Improve the pathological and histomorphological changes in mammary gland,the high dose group of haima could significantly increase the content of LH,the middle dose of haima significantly increased the content of LH,and the small dose of haima had no significant effect on LH.thymus,and spleen tissues of model mice.Conclusion:1.Itissuggestedthatthehaimacouldimprovedthebiochemical indicators,pathological parameters and appearance indicators of hyperplasia of mammary glands model animals in different degrees.2.Regulating the symptoms such as hyperplasia of mammary gland caused by disorder of estrogen level.3.It is found that reinforcing kidney yang medicine of haima can effectively treat hyperplasia of mammary gland,which lays a foundation for comprehensive utilization of hippocampus.
