| Mammary Gland Hyperplasia (MGH) is the first major disease in females, which cause and types are complicated, It mostly happens in women of 30-50 years in childbearing age. Its morbidity showing a gradually increasing and younger trend in recent years. It has been proved by pathological researches that the genesis of mammary carcinoma(MC)is closely related with MGH, and threated feminine physical and mental health. At present, Chinese Medicine has its own unique tradition strengths to treat MGH, and one of basic measure is regulate Gan-Qi and activate blood circulation to remove stasis,dredge collaterals, soften mass and lump.1. Literature and theoretical researchTo sum up physiological and pathological mechanism on MGH and its treatment overview. The exact mechanism of MGH genesis is not clear yet. According to modern medicine, the illness is related to endocrine maladjustment, and mainly due to ovary functional maladjustment disequilibrium of the ratio of progesterone to estrogen, i. e. decrease of excretion of progesterone and relative increase of estrogen affect excessive hyperplasia of mammary gland during the menstruation and incomplete recovery after the menstruation, which results in excessive stimulation of mammary tissues by estrogen, leading to hyperplasia of mammary dutal, lobular or the surrounding connective tissues cell. Viewing from modern medicine, regulating secretion ofthe pituitary in vivo-gonadal hormone , improving ovary function, regulating the disequilibrium level of hormone, and relieving clinical symptoms are considered the treatment key. To generalize the comprehension of TCM about etiology, pathogenesis and treatment of MGH. According to TCM, it belongs to the category of "Rupi","milk TB" and so on. Its main cause is closely related to the liver,spleen,Chong and Conception vessels, which pathogenesis of the disease characterists are " interal gloomy emotion,liver Qi stagnation; insufficiency of liver and kidney-the imbalance; phlegm and blood stasis condensed milk Tongxinluo blocked; Yangming Weire, thermal injury blood Tongxinluo" .To sum up treatment progress of TCM for MGH in recent years. Its basic principles of treatment are "regulate Gan-Qi and activate blood circulation to remove stasis and dredge collaterals, soften mass and lump" , the pharmacological study on DANSHEN,BIEJIA and seaweed illuminates theroitical basement of TCM on Danbie capsule.2 Experimental researches2. 1 ObjectiveTo study the effect of Danbie capsule on experimental Mammary Gland Hyperplasia and explore pharmacodynamics and its mechanism so that it can provide experimental basis for the development of a treatment drug for MGH and provide strong theoretical basis for further clinical research and clinical application.2.2 Methods(1) Study on effect of Danbie capsule on Experimental MGH model:①Effect of Danbie capsule on MGH in rabbits;②Effect of Danbie capsule on MGH in rats.70 female experimental creature were allocated and randomly divided into seven groups:the normal control group,the model group,three remedial groups with large, middle and small dosage of Danbie capsule,Rukuaixiao capsule group and Tamoxifen Citrate tablets group. Injecting diethylstilbestrol and estradiol benzoate induced the model of MGH, The levels of E2,T,P and PRL in plasma were determined, Changes of breast diameter and length, mammary gland volume and weight were measured, Pathological changes in morphology due to MGH were observed by microscopy, The organ coefficients were measured, too.(2)Effect of Danbie capsule on the experimental blood stasis creatural models:①Effect of Danbie capsule on blood stasis of microcirculation in mice②Effect of Danbie capsule on blood stasis of thrombosis in rats③Effect of Danbie capsule on blood stasis of blood rheology in rats. 70 experimental creature were allocated and randomly divided into seven groups: the normal control group,the model group,three remedial groups with large, middle and small dosage of Danbie capsule,Rukuaixiao capsule group and Tamoxifen Citrate tablets group. Injecting epinephrine hydrochloride subcutaneously and the ice bath induced the model of blood stasis, Changes of microcirculation of blood flow,whole blood viscosity, plasma viscosity,thrombosis in the wet weight and dry weight were measured.(3)Experimental investigation on effect of Danbie capsule on relieving pain:①Effect of acetic acid-induced pain(Writhing law)②Effect of heat-induced pain(Hot plate)2.3 Results(1) Study on effect of Danbie capsule on Experimental MGH model:The medicine can remrakly improve breast structure of MGH in rabbits and rats, reduce breast duct ectasia, ductal epithelial cell proliferation,the content of E2 and P,uterine index and the enlargement value of breast diameter and length, mammary gland volume and weight, middle dosage of Danbie capsule could also decrease ovarian index, with marked diference from the model group (P<0.05-0.01).(2)Effect of Danbie capsule on the experimental blood stasis creatural models:Danbie capsule can significantly speed up ear vascular blood flow on blood stasis of microcirculation in mice, with marked diference from the model group(P<0.01); reduce venous thrombosis in the wet weight and dry weight on blood stasis of blood rheology in rats, with marked diference from the model group(P<0.01), and acute blood stasis rat model of whole blood viscosity (low shear and high shear), small dosage may also reduce the plasma viscosity, Compared with the model group(P<0. 05-0. 01).(3) Experimental investigation on effect of Danbie capsule on relieving pain:when Acetic acid irritate ache of the mice, Danbie capsule can significantly relive the mice writhing response' s frequency as well as extend the time of writhing response coming, compared with the normal control group(P<0.05-0.01), It can Significantly enhanced the pain threshold by heat-induced pain in mice, compared with the normal control group(P<0.05-0.01).2.4 ConclusionDanbie capsule has the function of prevention and treatment action on experimental MGH, Its mechanism maybe adjust the ratio of estrogen and progesterone, improve the breast blood circulation, promote the absorption of pathological product, and elimination addiction analgesic effects.
